Desthiobiotin is a biotin derivative that retains the ability to bind tightly to streptavidin or avidin, but it lacks the biotin’s functional “H” group, making it a reversible or weaker binding interaction compared to biotin. This reduced affinity allows for the removal of the label under mild conditions, making Desthiobiotin useful in applications where temporary or reversible binding is needed. It is commonly used in Immunohistochemistry, protein purification, affinity chromatography, and Western Blotting. The desthiobiotin-avidin or desthiobiotin-streptavidin interaction can be employed for detection, capture, and separation of labeled biomolecules, offering greater flexibility in experimental designs that require label removal without affecting the underlying biomolecule’s functionality.
