Alkaline Phosphatase (AP)

Protein Detection and Visualization | Substrates: Alkaline Phosphatase (AP)

Alkaline Phosphatase (AP) is a robust and versatile enzyme employed extensively in immunohistochemistry (IHC), in situ hybridization (ISH), immunocytochemistry, and blotting applications. As provided by Vector Laboratories, AP functions optimally under alkaline conditions and catalyzes the hydrolysis of phosphate groups, enabling the visualization of biomolecular targets through substrate conversion. A defining advantage of AP lies in its independence from hydrogen peroxide, which not only reduces nonspecific background staining but also facilitates compatibility with peroxide-sensitive specimens. Vector Laboratories supplies a diverse array of AP substrates, including Vector® Red, Vector® Blue, Vector® Black, and BCIP/NBT, each yielding a distinct chromogenic or fluorescent precipitate. These substrates exhibit excellent stability and signal durability, even under dehydration and mounting conditions. The enhanced ImmPACT® Vector® Red substrate offers increased sensitivity for low-abundance targets. Owing to its low endogenous interference and compatibility with multiplex detection strategies, AP is particularly well-suited for sequential staining protocols and complex tissue environments where signal specificity and longevity are paramount.
