Azide-Modified Protein Capture using streptavidin-based enrichment is a technique that enables the selective isolation of azide-labeled proteins through Click Chemistry. In this approach, azide-modified proteins are reacted with biotin-alkyne probes via copper-catalyzed (CuAAC) or copper-free (SPAAC) click reactions, forming stable triazole linkages. The biotin-tagged proteins are then captured using streptavidin-coated magnetic beads or resin, allowing for efficient purification, enrichment, or downstream proteomic analysis.
