<p>The first image above is a composite image generated from individual images taken during six rounds of immunofluorescence staining using VectaPlex.  FFPE stomach; six mouse primary antibodies – CD20 (red), CD34 (yellow), Desmin (cyan), AE1/AE3 (magenta), CD3 (gray), Vimentin (green) – detected with DyLight™ 488 horse anti-mouse IgG.  Blue indicates DAPI stain.  Vector® TrueView® Autofluorescence Quencher Kit applied to suppress background fluorescence.</p>
<p>The second image above is a 4-plex IHC image taken of esophagus tissue stained for CD68 (brown) using ImmPACT DAB, CD20 (red) using ImmPACT Vector Red, CD34 (blue) using Vector Blue, and Desmin (grey) using ImmPACT SG.  Each round of staining was performed using normal IHC protocols with VectaPlex treatment between each round of staining.</p>
<p>The third image above was captured after three rounds of tyramide staining. VectaPlex was used after each round. FFPE tonsil is shown, following application of 3 mouse primary antibodies – CD3 (red), CD68 (green) and AE1/AE3 (purple) – detected using Opal 3-plex TSA Manual Detection Kit. Blue indicates DAPI stain.</p>

				        <h3>Technical Information</h3><p>This ready-to-use kit is designed to remove antibodies and other non-covalently bound detection reagents from formalin-fixed paraffin-embedded (FFPE) tissue sections in immunofluorescence or tyramide signal amplification staining workflows.</p><p>VectaPlex simplifies sample processing for manual workflows, accelerates multiplex assay optimization, preserves precious sample, and maintains tissue integrity and morphology through multiple cycles of antibody binding and removal.</p><p> </p><p> </p><p><b>VectaPlex IHC Workflow:</b> After staining with one of the approved substrates, VectaPlex is used to remove detection reagents in preparation for another cycle of staining. The substrate is not removed or impacted by VectaPlex. This reagent contains no animal-derived proteins and can be used as an alternative to sera, BSA, casein or non-fat dry milk in situations when animal protein-based blocking solutions cause increased background or interfere with blot development. Primary and secondary antibodies can be diluted in this solution and stored at 4 ºC for up to a week.</td>

		<p>Both products are derived from the same plant protein and in some applications can be used interchangeably. At a high level, one is presented as a ready-to-use (RTU) format and the other a more economical concentrate. The key difference is that the RTU format has been specifically optimized for use in IHC and IF applications. The RTU format has a neutral pH suitable for tissue and cell-based assays and has been more highly refined to avoid the presence of flocculent material that may cause interference with microscopy work. The 5x concentrated format is best suited for membrane blotting applications.</p>

				        <h3>Technical Information</h3><p>This plant protein derived product is a universal antibody diluent and blocking reagent intended for cell and tissue based immunohistochemistry (IHC) and immunofluorescent (IF) applications. This ready-to-use solution can be used as an alternative to normal sera, BSA, casein and non-fat dry milk in methods where these or other blocking agents have been found to be ineffective.</p><p>It is supplied without any sodium azide, and as such, can be used with both peroxidase and alkaline phosphatase antibody conjugates and all secondary detection reagents including polymer systems and avidin/biotin reagents that incorporate these enzymes. </p><p>Importantly, this is a different formulation than our concentrated (5x) animal free blocker, due to the optimized conditions and neutral pH of the solution for IHC and IF methods. </p>                    </div>

<p>Vector Laboratories fluorochrome-conjugated streptavidin and avidin reagents are highly purified and possess very low non-specific binding properties. They have extremely high affinity for biotin. These fluorescent conjugates can be used to detect biotinylated secondary antibodies and other macromolecules in applications such as immunofluorescence, in situ hybridization, or flow cytometry.</p>
<p>Amplification of fluorescent signals can be easily achieved with our biotinylated secondary antibodies followed by our highly purified fluorochrome-labeled streptavidin or avidin. These reagents are blended at optimized dilutions to provide a stable pre-diluted, ready-to-use HRP/AP formula that will detect mouse and rabbit primary antibodies, and facilitate a time-saving, simplified, one-step double label detection protocol. For maximum sensitivity and staining contrast between target antigens, ImmPACT DAB EqV (HRP, brown) and ImmPACT Vector Red (AP, magenta) substrates are included to complete this easy to use staining kit.</p><p>Reliable and reproducible results are obtained on tissue sections where the two target antigens do not over-lap (co-localize) in the same structure of the same cell, but rather are expressed in different cell compartments or different cell types.</p><p>Also included, BLOXALL Blocking Solution abolishes all endogenous peroxidase and alkaline phosphatase activity in a single 10 minute incubation. <p>The TrueVIEW Autofluorescence Quenching Kit provides a novel way to diminish unwanted autofluorescence from non-lipofuscin sources and dramatically improve signal-to-noise ratio. TrueVIEW removes unwanted autofluorescence in tissue sections due to aldehyde fixation, the presence of red blood cells, and structural elements such as collagen and elastin. The quenching action of the kit reagents provides a clear, unambiguous, “true view” localization of the target antigen.</p>

				        <h3>Description</h3><p>The TrueVIEW Autofluorescence Quenching Kit provides a novel way to diminish unwanted autofluorescence from non-lipofuscin sources and dramatically improve signal-to-noise ratio. TrueVIEW removes unwanted autofluorescence in tissue sections due to aldehyde fixation, the presence of red blood cells, and structural elements such as collagen and elastin. The quenching action of the kit reagents provides a clear, unambiguous, “true view” localization of the target antigen.</p><h3>This kit offers you:</h3><ul><li>Effectivity with problematic tissues such as kidney and spleen</li><li>An easy-to-use, one-step method</li><li>Compatibility with a wide selection of fluorophores</li><li>Compatibility with standard epifluorescence and confocal laser microscopes</li><li>Availability <a href="https://staging.vectorlabs.com/products/trueview-autofluorescence-quenching-kit-dapi">with DAPI counterstain</a> and without.</li></ul><p>One kit is sufficient to treat approximately 100 to 150 tissue sections. </p>                    </div>

		<p>During product development, applying the TrueVIEW™ Quenching reagent at the end of our standard IF procedure yielded the most optimal reduction in autofluorescence signal. The reagent is retained on the tissue at the time of mounting, allowing for extended quenching action, with little to no effect on the specific fluorescent signal. However, in applications with very brief staining procedures, such as a primary antibody directly conjugated with a fluorophore, application of TrueVIEW™ Quenching reagent may be just as effective at the start of the procedure.</p>

		<p>The working solution of TrueVIEW works in an electrostatic manner to greatly reduce or eliminate fluorescence in tissue sections induced through the use of an aldehyde based fixative. TrueVIEW is also effective at reducing fluorescence from tissue components such as collagen, elastin and red blood cells. TureVIEW does not work to reduce autofluorescence due to lipofuscin.</p>

		<p>Both TrueVIEW products, SP-8400 and SP-8500, are supplied with 2 mL of VECTASHIELD Vibrance antifade mounting media. We have found that the mounting media does play a crucial role in maintaining the high signal to noise ratio when using TrueVIEW. VECTASHIELD Vibrance is included therefore as a critical component of the kit and it is recommended to use the media supplied for optimal results. At this time we do not have sufficient data to confidently recommend the use of other vendors mounting media with TrueVIEW. Substitution of VECTASHIELD Vibrance with another mounting media may result in less than satisfactory results.</p>

The extent and intensity of autofluorescence background frequently makes it difficult or impossible to distinguish specific signals in immunofluorescence applications.</p><p>Most methods for reduction of tissue autofluorescence act primarily on lipofuscin granules, and are not broadly effective against the most common sources of autofluorescence targeted by TrueVIEW Quencher.</p><p>Current methods for reducing autofluorescence primarily include “home brew” concoctions such as sodium borohydride and other ink-based products. These methods are essentially ineffective against aldehyde induced autofluorescence. <p>The refractive index for VECTASHIELD Mounting Medium is 1.45.</p>
<p>VECTASHIELD PLUS Mounting Medium does not solidify, but rather, remains a liquid on the slide—which can then be stored without sealing. If desired, coverslips can be sealed around the perimeter using nail polish or plastic sealant. Mounted slides should be stored at 2-8 °C and protected from light.</p>
<h3><strong>VECTASHIELD Mounting Medium Antifade Comparison</strong></h3>
<p>Other manufacturers measure the antifade properties of their mountants using labeled microspheres or arrayed spots. Vector Labs prefers to measure antifade properties of VECTASHIELD mountants using frozen tissue sections that have been immunohistochemically stained with fluorescently labeled secondary antibodies. Antifade capability is then measured using a 40x objective with real time imaging over 30 seconds of continuous exposure to the excitation illumination. Individual intensity measurements are recorded from 6 separate labeled regions and the average is calculated. The intensity after 30 second exposure is expressed as a percentage of the intensity at zero time. The values for PG are taken from the manufacturer’s published results.</p>

				        <h3>Technical Information</h3><p>The refractive index for VECTASHIELD Mounting Medium is 1.45.</p><p>VECTASHIELD PLUS Mounting Medium does not solidify, but rather, remains a liquid on the slide—which can then be stored without sealing. If desired, coverslips can be sealed around the perimeter using nail polish or plastic sealant. Mounted slides should be stored at 2-8 °C and protected from light.</p><h3><strong>VECTASHIELD Mounting Medium Antifade Comparison</strong></h3><p>Other manufacturers measure the antifade properties of their mountants using labeled microspheres or arrayed spots. Vector Labs prefers to measure antifade properties of VECTASHIELD mountants using frozen tissue sections that have been immunohistochemically stained with fluorescently labeled secondary antibodies. Antifade capability is then measured using a 40x objective with real time imaging over 30 seconds of continuous exposure to the excitation illumination. Individual intensity measurements are recorded from 6 separate labeled regions and the average is calculated. The intensity after 30 second exposure is expressed as a percentage of the intensity at zero time. 				        <h3>Technical Information</h3><div class="product attribute description"><div class="value"><p>The H.O.H. Immunodetection Kit is intended to detect human (or humanized) antibodies on frozen or paraffin embedded human tissue sections.</p><p>The inability of a secondary antibody to distinguish between a primary antibody produced in humans and the endogenous human immunoglobulins present in human tissue results in high background staining, which obscures specific staining. This problem can be eliminated by using our Human on Human Kit and the result is clear, crisp, specific staining of the antigens of interest.<br /><br />This kit employs a straightforward two-step primary antibody preparation followed by standard IHC assay detection procedures. Once the human primary antibody solution has been prepared, assay time is approximately 90 minutes.</p><p>This comprehensive kit includes all required volume-matched reagents including <a href="https://staging.vectorlabs.com/products/immpact-dab-eqv-hrp-substrate">ImmPACT DAB EqV peroxidase substrate</a>. The procedures for use are easy to follow and omit the necessity of tedious and often confusing calculations. The H.O.H. Immunodetection Kit can be applied to most cell and tissue based staining situations where the detection of endogenous human IgG would be problematic when using a human, humanized, or chimeric human primary antibody.
