Amino-dPEG®24-OH, product number QBD-10868, also known as Amino-dPEG®24-alcohol, is a long (71 atoms, 86.0 Å), monodispersed PEGylation reagent designed to modify surfaces and biomolecules. One end of the molecule terminates with a primary amine, while the other end terminates with a primary alcohol group. This extremely hydrophilic PEGylation reagent reacts with carboxylate groups via the primary amine, leaving the hydroxy terminus free for functionalization and further reactions or as a hydrophilicity or charge modifier for the conjugate molecule.

The most common uses for Amino-dPEG®24-OH include modifying biomolecules and passivating surfaces with a highly hydrophilic coating. The primary amine on one end of the dPEG® linker reacts with carboxylates to form stable amide bonds and with aldehydes and ketones to form labile Schiff bases that can be reduced to secondary amines under mild conditions for greater stability. Conjugations with this molecule most commonly use the amine-carboxylate reaction with a carbodiimide such as EDC. Alternatively, activation of carboxylate as the NHS or TFP ester followed by reaction under slightly basic conditions also permits conjugation of the carboxylate and amino moieties.

Propargyl amine, product number 10510, is an amine-functionalized, three-carbon, terminal aliphatic alkyne. A non-dPEG® product, propargyl amine (formally, prop-2-yn-1-amine) is an alkynyl partner for Quanta BioDesign's azide-functionalized dPEG®-based click chemistry reagents. Click chemistry using propargyl amine and azides requires catalysis. Copper(I) catalysis leads to the 1,3-dipolar cycloaddition product. Ruthenium catalysis forms the 1,5-dipolar cycloaddition product. The terminal amine can be reacted with any of Quanta BioDesign's dPEG® NHS or TFP esters or carboxylic acid products to form any desired product.

A non-dPEG® product, propargyl amine (formally, prop-2-yn-1-amine) is an alkynyl partner for Quanta BioDesign’s azide-functionalized dPEG®-based click chemistry reagents. Click chemistry using propargyl amine and azides requires catalysis. Copper(I) catalysis leads to the 1,3-dipolar cycloaddition product. Ruthenium catalysis forms the 1,5-dipolar cycloaddition product. The terminal amine can be reacted with any of Quanta BioDesign’s dPEG® NHS or TFP esters or carboxylic acid products to form any desired product.

Difluorocarboxyfluorescein Cadaverine, 5-isomer (sold under Oregon Green™ 488 Cadaverine, 5-isomer name) is a carbonyl reactive building block used to modify carboxylic groups in the presence of activators (e.g. EDC, or DCC) or activated esters (e.g. NHS esters) through a stable amide bond. It also can be employed as a polar tracer and as a reactive dye for labeling proteins via a carboxylic acid moiety.

Difluorocarboxyfluorescein Cadaverineis a fluorinated analog of fluorescein that overcomes some of the key limitations of fluorescein, including greater photostability and a lower pKa (pKa ~ 4.7 versus 6.4 for fluorescein), making its fluorescence essentially pH insensitive in the physiological pH range.

Difluorocarboxyfluorescein Cadaverine, 5-isomer structurally is identical to Invitrogen’s Oregon Green™ 488 Cadaverine, 5-isomer

5-TAMRA cadaverine can used to modify carboxylic acid group in the presence of activators (e.g. EDC, or DCC) or activated esters (e.g. NHS esters) through a stable amide bond. It also can be reversibly coupled to aldehydes and ketones to form a Schiff base – which can be reduced to a generate stable amine derivative by sodium borohydride (NaBH4) or sodium cyanoborohydride (NaCNH3).

Although the mixed isomers of 5(6)-TAMRA cadaverine is a preferred, routinely used orange-fluorescent dye for staining proteins, it is rearly used for labeling peptides and nucleotides. Purification of 5(6)-TAMRA labeled peptide and nucleotides might be troublesome due to significant signal broadening in HPLC purification. Peptides and nucleotides labeled with a single isomer TAMRA usually give better resolution in HPLC purification that is often required in the conjugation processes.

AZDye™ 568 Cadaverine is a carbonyl reactive building block used to modify carboxylic groups in the presence of activators (e.g. EDC, or DCC) or activated esters (e.g. NHS esters) through a stable amide bond. It also can be employed as a polar tracer and as a reactive dye for labeling proteins via a carboxylic acid moiety.

AZDye™ 568 is a bright, and highly photostable, orange-fluorescent probe optimally excited by the 568 nm laser line on the Ar-Kr mixed-gas laser. This probe is water-soluble and its fluorescence is pH independent over a wide pH range. The brightness and photostability of this dye are best suited to direct imaging of low-abundance targets.

AZDye™ 405 Cadaverine is a carbonyl-reactive building block used to modify carboxylic groups in the presence of activators (e.g. EDC or DCC) or activated esters (e.g. NHS esters) through a stable amide bond. Another common application for AZDye™ 405 Cadaverine is cell fixing by treatment with formaldehyde or glutaraldehyde.

AZDye™ 405 is a water-soluble, blue-fluorescent dye that is often used in multi-color applications, including flow cytometry and super-resolution microscopy using STORM. Its excitation is ideally suited for the 407 nm spectral line of the krypton laser or the 408 nm violet laser diode. AZDye™ 405 is structurally and spectrally similar to DyLight® 405, Alexa Fluor® 405, CF® 405S Dye, or any other pyrene-based fluorescent dyes.

Cy5 Amine is a carbonyl reactive building block used to modify carboxylic groups in the presence of activators (e.g. EDC, or DCC) or activated esters (e.g. NHS esters) through a stable amide bond. It also can be employed as a polar tracer and as a reactive dye for labeling proteins via a carboxylic acid moiety.

Cy5 Amine (also sold under the Sulfo-Cyanine5 Amine name) is a water-soluble, bright, far-red-fluorescent probe with excitation ideally suited for the 633 nm or 647 nm laser lines. A significant advantage to using long wavelength dyes such as Cy5 or AZDye 647 over other fluorophores is the low autofluorescence of biological specimens in this region of the spectrum.
