<p>Tomato lectin, although sharing some specificities with potato lectin, Datura lectin, and wheat germ agglutinin, has been reported to be dissimilar in many respects. LEL binds well to glycophorin and Tamm-Horsfall glycoprotein and has been used effectively to label vascular endothelium in rodents.</p>
<p>Texas Red<sup>®</sup> labeled Tomato lectin has an appropriate number of fluorochromes bound to provide the optimum staining characteristics for this lectin. This conjugate is supplied essentially free of unconjugated fluorochromes. The excitation maximum is at 595 nm and the emission maximum is at 615 nm.</p>

		<p>The <em>Lycopersicon esculentum</em> (tomato) lectin has been widely reported as an effective blood vessel marker for <em>in vivo</em> vascular perfusion studies in rodent species. Investigators have primarily utilized one of the fluorophore conjugated tomato lectin formats to trace blood vasculature in normal and diseased animals via tail vein or intracardiac injection. However, the biotinylated format has also been used. It allows for flexibility in subsequent visualization by way of either fluorescence or enzyme-based methods. Published references are best source for procedural details. 				        <h3>Technical Information</h3><div class="product attribute description"><div class="value"><p>Tomato lectin is a very stable single subunit glycoprotein containing about 50 percent arabinose and galactose and may form multimeric aggregates in solution. </p><p>Tomato lectin (from <em>Lycopersicon esculentum</em>) is an effective marker of blood vessels and microglial cells in rodents. Conjugation of the lectin with a fluorophore facilitates fast, one-step detection and visualization using intravascular perfusion methods or direct application to tissue sections.</p><p>Texas Red<sup>®</sup> or DyLight<sup>™</sup> 594 conjugated tomato lectin provides an excellent contrast to green/yellow fluorescence such as GFP expressed in transgenic animals, or with fluorescein conjugates in standard double label studies. The tomato lectin complements our existing range of lectin reagents and should be a valuable tool in examining rodent tumor angiogenesis, tracing neovascular development in xenograft models and brain research.</p><p>Accompanying each fluorescent lectin is an analysis data sheet summarizing the results of our quality control tests and providing pertinent information on the product. These are then purified by an affinity chromatography procedure designed to remove any low affinity antibodies which may be present. The final product is then subjected to rigorous quality control assays including immunodiffusion, solid-phase enzyme immunoassays, gel electrophoresis and solid-phase binding assays. In preparing the labeled antibodies, great care is taken to ensure the maximum degree of labeling with no alteration in the specificity and affinity of the antibody. They have extremely high affinity for biotin. Amplification of fluorescent signals can be easily achieved with our biotinylated secondary antibodies followed by our highly purified fluorochrome-labeled streptavidin or avidin. Using a biotin/avidin or biotin/streptavidin detection system results in an additional layer of amplification over a directly conjugated secondary antibody.</p><p>Since the excitation and emission maxima are well separated from those of fluorescein, Texas Red Streptavidin can be employed with Fluorescein Streptavidin or other fluorescein conjugates to simultaneously localize two antigens in the same tissue section. <p>Avidin DCS, Texas Red<sup>®</sup> labeled, is a cell sorter grade of Texas Red<sup>®</sup> Avidin D. Through the development of a new method of labeling we are able to incorporate more fluorochrome into a protein without altering the properties of the protein. Not only does the Texas Red<sup>®</sup> Avidin DCS have a high F/P ratio, the efficiency of fluorescence is also increased. In addition to the improved conjugation technique, we employ a purification step to reduce nonspecific binding which can hamper resolution in cell sorting and other applications.</p>
