<p>VectaCell Trolox is an antifading additive for live cell imaging. It is a stock solution of the antioxidant Trolox (6-hydroxy-2,5,7,8-tetramethylchroman-2-carboxylic acid) in filtered ethanol. Trolox is a water-soluble and cell-permeable analog of vitamin E that efficiently prevents formation of various reactive oxygen species, such as singlet oxygen (O<sub>2</sub>), superoxide anion (O<sub>2</sub>) or hydrogen peroxide (H<sub>2</sub>O<sub>2</sub>).</p>

				        <h3>Technical Information</h3><div class="product attribute description"><div class="value"><p>VectaCell Trolox contains both Trolox and its oxidized form Trolox-quinone. This system reduces photo-bleaching and blinking during live cell imaging,<sup>1, 2</sup> and has also been shown to have a cytoprotective effect and low cytotoxicity for different cell lines.<sup>3</sup></p><p>Trolox has been used to reduce fluorophore bleaching<sup>1</sup> as well as the fluorophore blinking<sup>2</sup> during the observation of fluorescent labels. 				        <h3>Technical Information</h3><p>Wheat germ agglutinin (WGA) contains a group of closely related isolectins, with an isoelectric point about pH 9. The receptor sugar for WGA is <em>N</em>-acetylglucosamine, with preferential binding to dimers and trimers of this sugar. WGA can bind oligosaccharides containing terminal <em>N</em>-acetylglucosamine or chitobiose, structures which are common to many serum and membrane glycoproteins. Bacterial cell wall peptidoglycans, chitin, cartilage glycosaminoglycans, and glycolipids can also bind WGA. Native WGA has also been reported to interact with some glycoproteins via sialic acid residues (see succinylated WGA). This lectin is used for the purification of insulin receptors and for neuronal tracing.</p><p>WGA-HRP can be used for both anterograde and retrograde tracing. Our peroxidase-wheat germ agglutinin is specially prepared for neuronal transport studies. WGA-peroxidase features a high concentration of protein and is supplied in a 2 mg package at 40 mg/ml ready for injection. Unconjugated wheat germ agglutinin, also used in neuronal studies is available as 10 mg and 25 mg of salt-free, lyophilized lectin.</p><p>Inhibiting/Eluting Sugar: Chitin Hydrolysate or 500 mM <em>N</em>-acetylglucosamine with salt and/or acid elution generally required</p>                    </div>

<p><em>Phaseolus vulgaris</em> agglutinin is the name ascribed to a family of lectins, each of which consists of four subunits. There are two different types of subunits. One appears to be involved primarily in red cell agglutination and has been designated the “E” subunit (for erythroagglutinin). The other type is involved in lymphocyte agglutination and mitogenic activity and has been termed the “L” subunit (for leucoagglutinin). These subunits combine to produce five isolectins.</p>
<p>The unconjugated PHA-L has been found to be an excellent, specific marker for use in anterograde neuronal tracing</p>

If reconstituted in 1 ml of 10 mM phosphate, the resulting solution will be 5 mM phosphate, 10 mM NaCl with trace CaCl<sub>2</sub>. After reconstitution, and if appropriate for use, a preservative such as sodium azide (0.04%) may be added. If the lectin is to be used for mitogenic assays, filter solution through a sterile 0.22 µm filter immediately after reconstitution, aliquot and store frozen. NOTE: Mitogenic activity may be diminished by repeated freezing and thawing. Anterograde Neuronal Tracing: For neuronal transport studies, reconstitute in 0.2 ml of 10 mM sodium phosphate, pH 8.0.Both the pH and ionic strength of the reconstituting buffer are important for optimal anterograde transport. A specific protocol for this application is available on request.</td>

				        <h3>Technical Information</h3><div class="product-description-paragraph">PHA-L, with four L type subunits, does not agglutinate red cells but is a potent mitogen. The other three isolectins, designated E3L1, E2L2, and E1L3, have erythroagglutinating and mitogenic activities proportional to the number of respective E or L subunits. We have termed the mixture of the five isolectins PHA (E+L).</div><div class="product-description-paragraph"><p>After iontophoretic injection of unconjugated PHA-L, the approximate rate of anterograde transport is about 4-6 mm/day, with survival periods of over 18 days having been observed. Once transported, the unconjugated PHA-L is best visualized for light microscopy with antibody to the lectin, followed by the appropriate detection system and substrate. Fluorescent staining can also be used for a faster staining procedure.
