		<p>We offer a choice of either peroxidase or alkaline phosphatase based VECTASTAIN ABC kit detection systems. Selection of which enzyme system to use would be the first step. Most IHC applications involve detecting an unconjugated primary antibody. Consider the species in which the primary antibody is raised (e.g. rabbit), and then match to to corresponding species-specific VECTASTAIN ABC kit (e.g. VECTASTAIN ABC Rabbit IgG kit). If you already have a biotinylated target (i.e. biotinylated secondary antibody or primary antibody), then a standard VECTASTAIN ABC kit containing just the avidin/biotinylated enzyme reagents would be required. Note that only the VECTASTAIN Universal Elite PLUS ABC kit (PK-8200) contains a substrate.</p>

This ABC complex takes advantage of two important properties of avidin: 1) an extraordinarily high affinity for biotin (over one million times higher than antibody for most antigens), and 2) four biotin-binding sites. These properties allow macromolecular complexes (ABCs) to be formed by mixing Avidin DH (Reagent A) with its paired biotinylated enzyme (Reagent B) prior to use. The ABC reagent, once formed, remains stable for many hours after formation and can be used for several days after preparation.</p><p>The VECTASTAIN ABC Reagent can be used to detect any molecule that is biotinylated. This property gives the avidin-biotin complex (ABC) method great versatility in the types of targets that can be detected as well as the types of applications in which it can be employed. For example, to detect a primary antibody made in rabbit, the appropriate choice is a VECTASTAIN ABC Kit designated “Rabbit IgG”.</p><p>For additional versatility and convenience, Vector Laboratories offers secondary antibody products that recognize more than one species of primary antibody. Our Universal Biotinylated Anti-Mouse/Rabbit IgG secondary antibody (Cat. No. BA-1400) is a cocktail of biotinylated anti-mouse IgG and anti-rabbit IgG, designed for use with both rabbit and mouse primary antibodies. This antibody should not be used to stain tissues from rodents or rabbits due to its reactivity with IgG present in the tissues of these species. The Universal Pan-Specific Anti- Mouse/Rabbit/Goat secondary antibody (Cat. No. BA-1300) (which is included in our VECTASTAIN Universal Kits) is a cocktail of biotinylated anti-mouse IgG, anti-rabbit IgG, and anti-goat IgG that recognizes mouse, rabbit, and goat primary antibodies. This secondary antibody should not be used to stain rat, mouse, or other rodent, rabbit, goat, bovine, or sheep tissue due to potential reactivity with endogenous IgG.</p><h3>3) Choose a convenient format:</h3><p>Finally, several kits are available in prediluted, stabilized formats for convenience and ease of use. See kits designated R.T.U. or Ready-To-Use.</p><h3>Consider Species Cross-Reactivity</h3><p>When choosing the optimal detection system for your application, it is important to consider not only the species of the primary antibody but also the species of the tissue under examination. If the species of the primary antibody and the species of the tissue are closely related (e.g. rat and mouse), the biotinylated secondary antibody may cross react with endogenous IgG in the tissue section leading to background staining. The following three options minimize background staining in these instances:</p><ul><li>Biotinylate the primary antibody and detect with a Standard VECTASTAIN Elite ABC Kit, PK-6100 (only includes the avidin-biotin complex, or ABC reagent).</li><li>Use a biotinylated secondary antibody specifically adsorbed to remove cross-reacting antibodies of closely-related species (e.g. biotinylated anti-mouse IgG, rat adsorbed, Cat. 		<p>We offer a choice of either peroxidase or alkaline phosphatase based VECTASTAIN ABC kit detection systems. Selection of which enzyme system to use would be the first step. Most IHC applications involve detecting an unconjugated primary antibody. Consider the species in which the primary antibody is raised (e.g. rabbit), and then match to to corresponding species-specific VECTASTAIN ABC kit (e.g. VECTASTAIN ABC Rabbit IgG kit). If you already have a biotinylated target (i.e. biotinylated secondary antibody or primary antibody), then a standard VECTASTAIN ABC kit containing just the avidin/biotinylated enzyme reagents would be required. Note that only the VECTASTAIN Universal Elite PLUS ABC kit (PK-8200) contains a substrate.</p>

This ABC complex takes advantage of two important properties of avidin: 1) an extraordinarily high affinity for biotin (over one million times higher than antibody for most antigens), and 2) four biotin-binding sites. These properties allow macromolecular complexes (ABCs) to be formed by mixing Avidin DH (Reagent A) with its paired biotinylated enzyme (Reagent B) prior to use. The ABC reagent, once formed, remains stable for many hours after formation and can be used for several days after preparation.</p><p>The VECTASTAIN ABC Reagent can be used to detect any molecule that is biotinylated. This property gives the avidin-biotin complex (ABC) method great versatility in the types of targets that can be detected as well as the types of applications in which it can be employed. For example, to detect a primary antibody made in rabbit, the appropriate choice is a VECTASTAIN ABC Kit designated “Rabbit IgG”.</p><p>For additional versatility and convenience, Vector Laboratories offers secondary antibody products that recognize more than one species of primary antibody. Our Universal Biotinylated Anti-Mouse/Rabbit IgG secondary antibody (Cat. No. BA-1400) is a cocktail of biotinylated anti-mouse IgG and anti-rabbit IgG, designed for use with both rabbit and mouse primary antibodies. This antibody should not be used to stain tissues from rodents or rabbits due to its reactivity with IgG present in the tissues of these species. The Universal Pan-Specific Anti- Mouse/Rabbit/Goat secondary antibody (Cat. No. BA-1300) (which is included in our VECTASTAIN Universal Kits) is a cocktail of biotinylated anti-mouse IgG, anti-rabbit IgG, and anti-goat IgG that recognizes mouse, rabbit, and goat primary antibodies. This secondary antibody should not be used to stain rat, mouse, or other rodent, rabbit, goat, bovine, or sheep tissue due to potential reactivity with endogenous IgG.</p><h3>3) Choose a convenient format:</h3><p>Finally, several kits are available in prediluted, stabilized formats for convenience and ease of use. See kits designated R.T.U. or Ready-To-Use.</p><h3>Consider Species Cross-Reactivity</h3><p>When choosing the optimal detection system for your application, it is important to consider not only the species of the primary antibody but also the species of the tissue under examination. If the species of the primary antibody and the species of the tissue are closely related (e.g. rat and mouse), the biotinylated secondary antibody may cross react with endogenous IgG in the tissue section leading to background staining. 		<p>We offer a choice of either peroxidase or alkaline phosphatase based VECTASTAIN ABC kit detection systems. Selection of which enzyme system to use would be the first step. Most IHC applications involve detecting an unconjugated primary antibody. Consider the species in which the primary antibody is raised (e.g. rabbit), and then match to to corresponding species-specific VECTASTAIN ABC kit (e.g. VECTASTAIN ABC Rabbit IgG kit). If you already have a biotinylated target (i.e. biotinylated secondary antibody or primary antibody), then a standard VECTASTAIN ABC kit containing just the avidin/biotinylated enzyme reagents would be required. Note that only the VECTASTAIN Universal Elite PLUS ABC kit (PK-8200) contains a substrate.</p>

For example, to detect a primary antibody made in rabbit, the appropriate choice is a VECTASTAIN ABC Kit designated “Rabbit IgG”.</p><p>For additional versatility and convenience, Vector Laboratories offers secondary antibody products that recognize more than one species of primary antibody. Our Universal Biotinylated Anti-Mouse/Rabbit IgG secondary antibody (Cat. No. BA-1400) is a cocktail of biotinylated anti-mouse IgG and anti-rabbit IgG, designed for use with both rabbit and mouse primary antibodies. This antibody should not be used to stain tissues from rodents or rabbits due to its reactivity with IgG present in the tissues of these species. The Universal Pan-Specific Anti- Mouse/Rabbit/Goat secondary antibody (Cat. No. BA-1300) (which is included in our VECTASTAIN Universal Kits) is a cocktail of biotinylated anti-mouse IgG, anti-rabbit IgG, and anti-goat IgG that recognizes mouse, rabbit, and goat primary antibodies. This secondary antibody should not be used to stain rat, mouse, or other rodent, rabbit, goat, bovine, or sheep tissue due to potential reactivity with endogenous IgG.</p><h3>3) Choose a convenient format:</h3><p>Finally, several kits are available in prediluted, stabilized formats for convenience and ease of use. See kits designated R.T.U. or Ready-To-Use.</p><h3>Consider Species Cross-Reactivity</h3><p>When choosing the optimal detection system for your application, it is important to consider not only the species of the primary antibody but also the species of the tissue under examination. If the species of the primary antibody and the species of the tissue are closely related (e.g. rat and mouse), the biotinylated secondary antibody may cross react with endogenous IgG in the tissue section leading to background staining. The following three options minimize background staining in these instances:</p><ul><li>Biotinylate the primary antibody and detect with a Standard VECTASTAIN Elite ABC Kit, PK-6100 (only includes the avidin-biotin complex, or ABC reagent).</li><li>Use a biotinylated secondary antibody specifically adsorbed to remove cross-reacting antibodies of closely-related species (e.g. biotinylated anti-mouse IgG, rat adsorbed, Cat. 		<p>A commonly used negative control is omission of the primary antibody. While this control addresses whether the secondary antibody reagents are a source of staining, inadvertent binding of the primary antibody to the tissue can occur. Various tissue elements such as Fc receptors and charged molecules may bind the primary antibody non-specifically. Simply omitting the primary antibody as a negative control would miss potential false positive staining by this means. A few “publication worthy” negative controls for IHC are listed below: 1) Preabsorption of primary antibody with the immunogen used to generate the antibody can be employed. The working dilution of the primary antibody and an optimized concentration of the immunogen are incubated together for a period prior to application to the specimen. Lack of staining would indicate specificity of the primary antibody to the target antigen in solution. Positive staining using this method, however, may indicate lack of specificity of the primary antibody and/or the primary antibody is being bound by tissue elements. To rule out the latter, this control can be used in combination with suggestion no. 2 below. Note that adsorption controls are not always feasible or practical depending on the cost or source of the immunogen. 2) Use of an isotype control (e.g. “non-immune” mouse IgG), matched to that of the primary antibody and applied at the same protein concentration as the primary antibody, is probably the most widely used negative control. This control addresses whether tissue elements are inadvertently binding immunoglobulin from the same species as the primary antibody, in addition to non-specific binding from the secondary detection reagents. In most cases, use of a sub-class of isotype immunoglobulin (e.g. mouse IgG2a or IgG2b) is not required. Note that the use of pre-immune immunoglobulin, obtained prior to immunization, could also be used. However, it is very unusual for commercial vendors to offer pre-immune immunoglobulin. 3) Substitution of the primary antibody with an “irrelevant antibody” is also a suitable negative control. The term “irrelevant” refers to a primary antibody of the same isotype as the specific primary antibody (i.e. mouse IgG) and applied at the same concentration, that is known not to bind to a target in the tissue specimen. An example would be an anti-cytokeratin antibody on smooth muscle tissue. As with negative control no. 1 above, lack of staining indicates tissue elements are not binding this isotype of immunoglobulin. 4) In some cases, the target antigen can be removed from the tissue specimen as a sort of “knock-out” preparation. Once the target antigen has been removed, the complete assay is run to determine lack of staining. One method to remove the target antigen is using defined enzyme digestion. Examples include the use of a collagenase if the target antigen is collagen, or hyaluronidase if the target antigen is hyaluronic acid. Of course, there are limitations to this approach, however, variations of this “deletion” or “knock-out” approach would be valid negative controls.</p>

<p>The rabbit anti-goat IgG antibodies are prepared by hyperimmunizing animals in a manner that produces high affinity antibodies. These are then purified by an affinity chromatography procedure designed to remove any low affinity antibodies which may be present. Cross-reactivities that are likely to interfere with specific labeling are removed by solid-phase adsorption techniques. The final product is then subjected to rigorous quality control assays including immunodiffusion, solid-phase enzyme immunoassays, gel electrophoresis and solid-phase binding assays. In preparing the labeled antibodies, great care is taken to ensure the maximum degree of labeling with no alteration in the specificity and affinity of the antibody. The labeled antibody then undergoes a further series of quality control assays, including immunohistochemical analysis.</p>
<p>AMCA (7-amino-4-methylcoumarin-3-acetic acid) labeled Rabbit Anti-Goat IgG Antibody excites in the near ultraviolet (350 nm) and emits a visible bright blue fluorescence at about 450 nm. This antibody has been optimally labeled with AMCA to provide the brightest label for fluorescence microscopy.</p>
<p>The use of bovine products such as casein, serum, albumin or non-fat dry milk as blocking agents in ELISA or blot assays may produce a high background due to cross-reactivity of this antibody with bovine immunoglobulins that may be present. If this antibody is to be used in these applications, blocking with Animal-Free Blocker<sup>®</sup> solution (Cat. No. SP-5030) is recommended.</p>

<p>The biotinylated rabbit anti-sheep IgG secondary antibody is used in an avidin-biotin or streptavidin-biotin detection system. These antibodies can be used for tissue and cell staining, ELISAs, and blots.</p>

<td class="col data" data-th="Recommended Usage">The recommended concentration range for use is 2-10 µg/ml. If this biotinylated antibody is to be used in tissues, which may contain cross-reacting endogenous immunoglobulins, dilution of this biotinylated antibody may be made in buffers containing 2% normal serum from the same species as the tissue. Important Note: The use of bovine products such as casein, serum, albumin or non-fat dry milk as blocking agents in ELISA or blot assays may produce a high background due to cross-reactivity of anti-sheep IgG with bovine immunoglobulins that may be present. If this antibody is to be used in these applications, blocking with Animal-Free Blocker™ solution (Cat. No. SP-5030) is recommended. Note:This product is at the same concentration as that provided in the VECTASTAIN® ABC kits.</td>

<p>Vector Laboratories affinity-purified antibodies are of unmatched quality. These antibodies are prepared using proprietary immunization schedules that produce high affinity antibodies. The antibodies are then purified by affinity chromatography, and cross-reactivities that are likely to interfere with specific labeling are removed by solid phase adsorption techniques. The biotinylated secondary antibodies are conjugated to ensure the maximum degree of labeling without compromising the specificity or affinity of the antibody.</p>
<p>With some exceptions the recommended dilution for most applications is 1:200.</p>
<p>The use of bovine products such as casein, serum, albumin or non-fat dry milk as blocking agents in ELISA or blot assays may produce a high background due to cross-reactivity of this antibody with bovine immunoglobulins that may be present.
