<p>Highly active WestVision™ Peroxidase Polymer antibody conjugates are western blot detection reagents. These reagents are:</p>

<p>WestVision™ Peroxidase Polymer antibody conjugates bind primary antibodies of mouse (WB-2000) or rabbit (WB-1000) origin with high affinity, producing sharp signal and low background in western blot detection. The conjugates are based on a new method of polymerizing enzymes and attaching these polymers to antibodies. The novel approach employed to form enzyme polymers avoids the use of large dextrans or other macromolecules as a backbone. Attaching these unique polymers to an antibody allows a higher density of enzymes to access a target with minimal interference. The result is an excellent signal-to-noise-ratio in western blot detection.</li>

<p>The DuoLuX Chemiluminescent/Chemifluorescent Substrate can be used for western, Southern, northern, or dot blots, or for ELISA applications. This substrate is about 10 times more sensitive than the alkaline phosphatase substrate BCIP/NBT and about 100 times more sensitive than the peroxidase substrate DAB. In addition to its chemiluminescent properties, the reaction product of the DuoLuX Substrate is also fluorescent.</p>

Reagents 1 and 2 are mixed in equal volumes just prior to use, providing 200 ml of peroxidase substrate working solution.</p><h3>Chemiluminescent properties</h3><p>Light emission of the DuoLuX Chemiluminescent/ Chemifluorescent Substrate occurs continuously over a long period of time for both the peroxidase and alkaline phosphatase substrates. With either substrate formula, reacted DuoLuX Substrate luminesces in the blue range with a peak emission at 453 nm. Fluorescence can be recorded with a digital imaging system or a conventional camera months after chemiluminescence has faded. The excitation maximum is at 405 nm, but other wavelengths (254 nm and 365 nm) also excite. Maximum fluorescent emission occurs at 453 nm. For fluorescence detection, nitrocellulose is recommended due to the intrinsic fluorescence of nylon. (Same 1 second acquisition time.)</em></p><h3>Applications</h3><p>The DuoLuX Chemiluminescent/Chemifluorescent Substrate is ideal for development of protein and nucleic acid blots.  Complete kits are available for western blot development (VECTASTAIN<sup>®</sup> ABC-AmP system).</p><h3>Protein Detection</h3><p>For western For western blot applications using chemiluminescent detection, the sensitivity using either alkaline phosphatase or peroxidase is approximately 1 pg of protein. Film exposure times using peroxidase are generally 5-30 seconds; exposure times for alkaline phosphatase are 1-5 minutes. Using fluorescence detection, sensitivities are less than 10 pg of protein. Using peroxidase, exposure times are somewhat shorter and sensitivities are slightly less than those for alkaline phosphatase. 				        <h3>Technical Information</h3><p>Wheat germ agglutinin (WGA) contains a group of closely related isolectins, with an isoelectric point about pH 9. The receptor sugar for WGA is <em>N</em>-acetylglucosamine, with preferential binding to dimers and trimers of this sugar. WGA can bind oligosaccharides containing terminal <em>N</em>-acetylglucosamine or chitobiose, structures which are common to many serum and membrane glycoproteins. Bacterial cell wall peptidoglycans, chitin, cartilage glycosaminoglycans, and glycolipids can also bind WGA. Native WGA has also been reported to interact with some glycoproteins via sialic acid residues (see succinylated WGA). This lectin is used for the purification of insulin receptors and for neuronal tracing.</p><p>WGA-HRP can be used for both anterograde and retrograde tracing. Our peroxidase-wheat germ agglutinin is specially prepared for neuronal transport studies. WGA-peroxidase features a high concentration of protein and is supplied in a 2 mg package at 40 mg/ml ready for injection. Unconjugated wheat germ agglutinin, also used in neuronal studies is available as 10 mg and 25 mg of salt-free, lyophilized lectin.</p><p>Inhibiting/Eluting Sugar: Chitin Hydrolysate or 500 mM <em>N</em>-acetylglucosamine with salt and/or acid elution generally required</p>                    </div>

				        <h3>Technical Information</h3><p>Our high affinity Horse Anti-Goat IgG Antibody is purified by affinity chromatography and adsorbed to remove any cross-reactivities that are likely to interfere with specific labeling. This antibody is then cross-linked with the highest specific activity horseradish peroxidase. A proprietary procedure for conjugation allows the maximum preservation of enzyme activity and antibody specificity. This antibody is subjected to rigorous quality control assays and can be used for tissue staining, ELISAs, and blots.</p><p>As well as recognizing goat IgG, this antibody is suitable to detect sheep and bovine IgG primary antibodies.</p><p>The use of bovine products such as casein, serum, albumin or non-fat dry milk as blocking agents in ELISA or blot assays may produce a high background due to cross-reactivity of this antibody with bovine immunoglobulins that may be present. If this antibody is to be used in these applications, blocking with Animal-Free Blocker<sup>®</sup> solution (Cat. No. SP-5030) is recommended.</p>                    </div>

This adsorbed antibody allows primary antibodies made in rat to be used on mouse tissues.</p><p>Peroxidase Goat Anti-Rat IgG, mouse adsorbed, is supplied in solution. With some exceptions, working concentration of HRP Anti-Rat IgG, mouse adsorbed, is 0.5-5.0 μg/ml.</p>                    </div>
