m-dPEG®25-TFP ester, product number QBD-11290, is a long (77 atoms, 89.9 Å), methyl-terminated, discrete-chain-length polyethylene glycol (dPEG®) spacer functionalized with a 2,3,5,6-tetrafluorophenyl (TFP) ester for reaction with free amines. m-dPEG®25-TFP ester is just one member of an comprehensive line of methyl-terminated PEGylation reagents that includes dPEG® spacers containing 2 to 49 ethylene glycol units.

TFP esters react optimally with free amines at pH 7.5 – 8.0 and are generally more hydrolytically stable in water and aqueous buffers than NHS esters. In aqueous media, the hydrolytic rate of the ester to the carboxylic acid increases with increasing pH. Reacting surface amines on biomolecules (e.g., proteins and peptides) with this uncharged, methyl-capped dPEG® spacer may alter the overall charge of the resulting conjugates.

Many applications could employ m-dPEG®25-TFP ester, including the following:
dendrimer construction;
PK and immunogenicity improvements for dendrimers, peptides, and proteins;
cell surface engineering;
peptide synthesis and modification to improve water solubility or decrease immunogenicity;
coating of nanoparticles, quantum dots, and carbon nanotubes; and
prevention of protein aggregation.

m-dPEG®12-TFP ester, product number QBD-10306, is a medium-length (38 atoms, 44.0 Å), methyl-terminated, discrete-length polyethylene glycol (dPEG®) spacer. With a reactive end of 2,3,5,6-tetrafluorophenyl (TFP) ester, m-dPEG®12-TFP ester is designed to modify surfaces having accessible free amines.

Free amines react optimally with TFP esters in aqueous media at pH 7.5 – 8.0. TFP esters are more hydrolytically stable in water and aqueous buffers than NHS esters. Nevertheless, in aqueous media, the rate at which the ester hydrolyzes to the carboxylic acid increases as the pH increases.
There are numerous applications that could successfully employ m-dPEG®12-TFP ester, including the following:
vaccine development;
cell surface engineering;
imaging applications to reduce background and non-specific interactions;
construction of dendrimers;
coating of nanoparticles, quantum dots, and carbon nanotubes;
improving PK and BD of biomolecules; and,
preventing enzyme aggregation.

m-dPEG®24-TFP ester, product number QBD-10303, is a long (74 atoms, 86.2 Å), methyl-terminated, discrete-length polyethylene glycol (dPEG®) spacer. With a reactive end of 2,3,5,6-tetrafluorophenyl (TFP) ester, m-dPEG®24-TFP ester is designed to modify surfaces having accessible free amines.

Moreover, it should be noted that reacting surface amines on biomolecules (e.g., proteins and peptides) with this uncharged, methyl-capped dPEG® spacer may alter the overall charge of the resulting conjugates.

Many applications could potentially use m-dPEG®24-TFP ester successfully. Among those applications are:
cell surface engineering;
PK, BD, and immunogenicity profile improvements for biomolecules;
imaging applications by reducing background and non-specific binding;
dendrimer construction;
nanoparticle, quantum dot, and carbon nanotube surface coating; and,
prevention of protein aggregation.

"m-dPEG®12-amido-dPEG®12-TFP ester, product number QBD-10149, is a long (77 atoms, 91.3 Å), methyl-terminated, discrete-length polyethylene glycol (dPEG®) spacer. The reactive end of the molecule is a 2,3,5,6-tetrafluorophenyl (TFP) ester. An amide bond joining two discrete PEG subunits is in the molecule's center. m-dPEG®12-amido-dPEG®12-TFP ester is designed to modify surfaces containing accessible free amines. Applications for m-dPEG®12-amido-dPEG®12-TFP ester include cell surface engineering, improving PK/BD, reducing bioconjugate immunogenicity, preventing protein aggregation, constructing dendrimers, and many more.

m-dPEG®12-amido-dPEG®12-TFP ester reacts with amine-functionalized surfaces (carbon nanotubes, nanoparticles made of silica or various metals, quantum dots, and similar types of products) or free amines on biomolecules. When used to coat surfaces or modify biomolecules, m-dPEG®12-amido-dPEG®12-TFP ester reduces or eliminates non-specific binding and increases water solubility. Crucially, the elimination of non-specific binding may increase the signal-to-noise ratio in imaging applications and assays where this is a consideration. Please note that modifying the surface amines of biomolecules with this uncharged, methyl-capped dPEG® spacer may alter the overall charge of the resulting conjugates. When reacting TFP esters with free amines, the optimal pH range is 7.5 - 8.5."

The reactive end of the molecule is a 2,3,5,6-tetrafluorophenyl (TFP) ester. An amide bond joining two discrete PEG subunits is in the molecule’s center. m-dPEG®12-amido-dPEG®12-TFP ester is designed to modify surfaces containing accessible free amines.

When reacting TFP esters with free amines, the optimal pH range is 7.5 – 8.5.

m-dPEG®48-NH-CO(CH2)3CO-TFP ester, product number QBD-10143, is a long (151 atoms, 178.5 Å), methyl-terminated, discrete-length polyethylene glycol (dPEG®) spacer. With a reactive end of 2,3,5,6-tetrafluorophenyl (TFP) ester, m-dPEG®48-NH-CO(CH2)3CO-TFP ester is designed to modify surfaces containing available free amines.

In aqueous media, free amines react optimally with TFP esters at pH 7.5 – 8.5. Compared to N-hydroxysuccinimidyl (NHS) esters, TFP esters are more hydrolytically stable in water and aqueous buffer. Nevertheless, in aqueous media, the rate at which the ester hydrolyzes to the carboxylic acid increases as the pH rises. Moreover, please note that reacting free amines on biomolecules with this uncharged, methyl-capped dPEG® spacer may alter the overall charge of the resulting conjugates.

Many applications potentially could use m-dPEG®48-NH-CO(CH2)3CO-TFP ester successfully. Among those applications are: cell surface engineering; PK, BD, and immunogenicity profile improvements for biomolecules; imaging applications, by reducing background and non-specific binding and consequently increasing the signal-to-noise ratio; dendrimer construction; nanoparticle, quantum dot, and carbon nanotube surface coating; and, prevention of protein aggregation."
