m-dPEG®25-NHS ester, product number QBD-11291, is a long (77 atoms, 89.9 Å), methyl-terminated, discrete-chain-length polyethylene glycol (dPEG®) spacer functionalized with an N-hydroxysuccinimidyl (NHS) ester for reaction with free amines. m-dPEG®25-NHS ester is just one member of an comprehensive line of methyl-terminated PEGylation reagents that includes dPEG® spacers containing 2 to 49 ethylene glycol units.

NHS esters react optimally with free amines at pH 7.0 – 7.5. In aqueous media, the hydrolytic rate of the ester to the carboxylic acid increases with increasing pH. Reacting surface amines on biomolecules (e.g., proteins and peptides) with this uncharged, methyl-capped dPEG® spacer may alter the overall charge of the resulting conjugates.

Many applications could employ m-dPEG®25-NHS ester, including the following:
dendrimer construction;
PK and immunogenicity improvements for dendrimers, peptides, and proteins;
cell surface engineering;
peptide synthesis and modification to improve water solubility or decrease immunogenicity;
coating of nanoparticles, quantum dots, and carbon nanotubes; and
prevention of protein aggregation.

m-dPEG®25-TFP ester, product number QBD-11290, is a long (77 atoms, 89.9 Å), methyl-terminated, discrete-chain-length polyethylene glycol (dPEG®) spacer functionalized with a 2,3,5,6-tetrafluorophenyl (TFP) ester for reaction with free amines. m-dPEG®25-TFP ester is just one member of an comprehensive line of methyl-terminated PEGylation reagents that includes dPEG® spacers containing 2 to 49 ethylene glycol units.

TFP esters react optimally with free amines at pH 7.5 – 8.0 and are generally more hydrolytically stable in water and aqueous buffers than NHS esters.

Many applications could employ m-dPEG®25-TFP ester, including the following:
dendrimer construction;
PK and immunogenicity improvements for dendrimers, peptides, and proteins;
cell surface engineering;
peptide synthesis and modification to improve water solubility or decrease immunogenicity;
coating of nanoparticles, quantum dots, and carbon nanotubes; and
prevention of protein aggregation.

m-dPEG®25-acid, product number QBD-11289, is a long (77 atoms, 89.9 Å), methyl-capped, discrete chain length polyethylene glycol (dPEG®) spacer that terminates in a propionic acid group. The terminal propionic acid moiety can be coupled directly to free amines using EDC or another carbodiimide. Alternatively, the reactive end can be converted into an active ester that can then be conjugated to free amines. N-hydroxysuccinimide (NHS), 2,3,5,6-tetrafluorophenol (TFP), and 2,3,4,5,6-pentafluorophenol (PFP) typically are used for this purpose.

m-dPEG®25-acid can modify amine-functionalized surfaces (carbon nanotubes, other nanoparticles, quantum dots, etc.) or free amines on biomolecules. When used to coat surfaces or modify biomolecules, m-dPEG®25-acid reduces or eliminates non-specific binding, increases the hydrodynamic volume and water solubility of the conjugate molecule. Please note that modifying the surface amines of biomolecules with this uncharged, methyl-capped dPEG® spacer may alter the overall charge of the resulting conjugates.

m-dPEG®25-amido-dPEG®24-acid, product number QBD-11097, is a long (153 atoms, 179.8 Å), methyl-capped, discrete chain length polyethylene glycol (dPEG®) spacer. The effective length of the dPEG® spacer is about 49 ethylene oxide units (i.e., m-dPEG®49-acid) with an amide bond in the middle of the chain. The reactive end of the molecule terminates in a propionic acid group. The terminal propionic acid moiety can be coupled directly to free amines using EDC or another carbodiimide. Alternatively, converting the reactive end into an active ester permits the conjugation of the transformed product to free amines. N-hydroxysuccinimide (NHS), 2,3,5,6-tetrafluorophenol (TFP), and 2,3,4,5,6-pentafluorophenol (PFP) are typically used for this purpose.

m-dPEG®25-amido-dPEG®24-acid, QBD-11097, reacts with amine-functionalized surfaces (carbon nanotubes, nanoparticles made of silica or various metals, quantum dots, and similar types of products) or free amines on biomolecules. When used to coat surfaces or modify biomolecules, m-dPEG®25-amido-dPEG®24-acid reduces or eliminates non-specific binding and increases water solubility. Crucially, the elimination of non-specific binding may increase the signal-to-noise ratio in imaging applications and assays where this is a consideration. Please note that modifying the surface amines of biomolecules with this uncharged, methyl-capped dPEG® spacer may alter the overall charge of the resulting conjugates.

Description

m-dPEG®25-amido-dPEG®24-acid, product number QBD-11097, is a long (153 atoms, 179.8 Å), methyl-capped, discrete chain length polyethylene glycol (dPEG®) spacer.

Please note that modifying the surface amines of biomolecules with this uncharged, methyl-capped dPEG® spacer may alter the overall charge of the resulting conjugates.

m-dPEG®25-amido-dPEG®24-TFP ester, product number QBD-11096, is a long (153 atoms, 178.9 Å), methyl-terminated, discrete-chain-length polyethylene glycol (dPEG®) spacer functionalized with a 2,3,5,6-tetrafluorophenyl (TFP) ester for reaction with free amines. It is effectively a single molecular weight mPEG49 linker with an amide linkage in the middle of the discrete PEG chain. m-dPEG®25-amido-dPEG®24-TFP ester is just one member of an comprehensive line of methyl-terminated PEGylation reagents that includes dPEG® spacers containing 2 to 49 ethylene glycol units.

TFP esters react optimally with free amines at pH 7.5 – 8.0 and are generally more hydrolytically stable in water and aqueous buffers than NHS esters.

Many applications could employ m-dPEG®25-amido-dPEG®24-TFP ester, including the following:
dendrimer construction;
PK and immunogenicity improvements for dendrimers, peptides, and proteins;
cell surface engineering;
peptide synthesis and modification to improve water solubility or decrease immunogenicity;
coating of nanoparticles, quantum dots, and carbon nanotubes; and
prevention of protein aggregation.

m-dPEG®13-TFP ester, product number QBD-11087, is a medium-length (41 atoms, 47.3 Å), methyl-terminated, discrete-chain-length polyethylene glycol (dPEG®) spacer functionalized with a 2,3,5,6-tetrafluorophenyl (TFP) ester for reaction with free amines. m-dPEG®13-TFP ester is just one member of an comprehensive line of methyl-terminated dPEG® products that includes dPEG® spacers containing 2 to 49 ethylene glycol units.

TFP esters react optimally with free amines at pH 7.5 – 8.0 and are generally more hydrolytically stable in water and aqueous buffers than NHS esters.

Many applications could employ m-dPEG®13-TFP ester, including the following:
vaccine development (modifying the immunogenicity of biomolecules);
dendrimer construction;
PK and immunogenicity improvements for dendrimers, peptides, and proteins;
cell surface engineering;
peptide synthesis and modification to improve water solubility or decrease immunogenicity;
coating of nanoparticles, quantum dots, and carbon nanotubes; and
prevention of protein aggregation.

m-dPEG®13-acid, product number QBD-11085, is a medium-length (41 atoms, 48.5 Å), methyl-capped, discrete chain length polyethylene glycol (dPEG®) spacer that terminates in a propionic acid group. The terminal propionic acid moiety can be coupled directly to free amines using EDC or another carbodiimide. Alternatively, the reactive end can be converted into an active ester that can then be conjugated to free amines. N-hydroxysuccinimide (NHS), 2,3,5,6-tetrafluorophenol (TFP), and 2,3,4,5,6-pentafluorophenol (PFP) typically are used for this purpose.

m-dPEG®13-acid can modify amine-functionalized surfaces (carbon nanotubes, other nanoparticles, quantum dots, etc.) or free amines on biomolecules. When used to coat surfaces or modify biomolecules, m-dPEG®13-acid reduces or eliminates non-specific binding, increases the hydrodynamic volume and water solubility of the conjugate molecule. Please note that modifying the surface amines of biomolecules with this uncharged, methyl-capped dPEG® spacer may alter the overall charge of the resulting conjugates.

m-dPEG®23-OH, product number QBD-10942, is a chemical modification reagent consisting of a long (70 atoms, 83.1 Å), methyl-terminated, water-soluble, non-immunogenic, single molecular weight, discrete polyethylene glycol (dPEG®) chain that terminates with a reactive primary alcohol (i.e., a hydroxy group). The hydroxy group can be functionalized with different groups that enable various types of reactions.

This product is comparable to mPEG1000, but it is a monodispersed PEG rather than a heterogeneous polymer PEG. Functionalization of the terminal alcohol with a variety of different groups permits this product to bring the benefits of dPEG® products to surfaces, small molecules, and biomolecules, including peptides, proteins, carbohydrates, and oligonucleotides. One useful application of this product is in coating surfaces to prevent non-specific or hydrophobic interactions. Other benefits that m-dPEG®23-OH brings to conjugated molecules include increased hydrodynamic volume (leading to reduced renal clearance), better water solubility, and reduced antigenicity.

Other benefits that m-dPEG®23-OH brings to conjugated molecules include increased hydrodynamic volume (leading to reduced renal clearance), better water solubility, and reduced antigenicity.

m-dPEG®48-MAL, product number QBD-10932, is a sulfhydryl-reactive, single molecular weight, with a long (152 atoms, 178.6 Å), discrete-chain-length PEG (dPEG®). This product modifies surfaces and biomolecules with free thiol groups through the maleimide reactive group. The non-immunogenic, methyl-terminated dPEG® chain has no charge but adds water solubility and hydrodynamic volume to molecules to which it is conjugated.

The reaction of the maleimide end of m-dPEG®48-MAL, product number QBD-10932, with a sulfhydryl proceeds best at pH 6.5 – 7.5. Conduct the conjugation at the lowest reasonable pH within this range. Above pH 7.5, free amines compete with free thiols at the maleimide reaction site, which creates confusing results. Moreover, at higher pH values, the maleimide ring may open to form unreactive maleamic acid.

Possible uses for m-dPEG®48-MAL include the following:
Surface coating and passivation of thiolated nanoparticles;
Modification of cysteine residues on peptides, proteins, and other sulfhydryl-containing molecules to increase hydrophilicity and decrease immunogenicity;
Modification of cysteine residues on peptides, proteins, and other sulfhydryl-containing molecules to increase hydrodynamic volume and reduce or eliminate renal clearance of the modified compound;
Supramolecular construction (for example, of hydrogels); and,
As a probe of protein structure or enzymatic function.
