The peroxidase micropolymers of the ImmPRESS HRP polymer reagent limit steric interference and provide enhanced accessibility to the target, avoiding the disadvantages of other polymer systems that use large dextrans or other macromolecules as backbones. The result is crisp, strong staining of antibody targets, especially nuclear and membrane antigens (such as Ki67, estrogen receptor, bcl-2, CD3, CD8 and CD10) and greater sensitivity than other polymer systems.

Staining Procedure

The staining procedure is simple as shown in the diagram below. Following a blocking step with the diluted normal horse serum, sections are incubated with primary antibody. After a brief wash, the appropriate ImmPRESS Reagent is added to the sections and incubated for 30 minutes. Sections are again rinsed and the slides are developed with the peroxidase substrate of choice.

Consider Species Cross-Reactivity

When choosing the optimal detection system for your application, it is important to consider not only the species of the primary antibody but also the species of the tissue. If the species of the primary antibody and the species of the tissue are closely related (e.g. rat and mouse), the secondary antibody may bind to endogenous IgG in the tissue section leading to background. The following options minimize background staining in these instances:

  • Use a secondary antibody specifically adsorbed to remove cross-reacting antibodies of closely-related species (e.g. ImmPRESS Anti-Mouse IgG, Rat Adsorbed).
  • Use the M.O.M.® ImmPRESS Kit (Cat. No. MP-2400) for applications of mouse primary antibodies on mouse tissue.

Technical Information

Save time and streamline your double staining IHC workflow with the ImmPRESS Duet Double Staining Polymer Kit. These reagents are blended at optimized dilutions to provide a stable pre-diluted, ready-to-use HRP/AP formula that will detect mouse and rabbit primary antibodies, and facilitate a time-saving, simplified, one-step double label detection protocol. For maximum sensitivity and staining contrast between target antigens, ImmPACT DAB EqV (HRP, brown) and ImmPACT Vector Red (AP, magenta) substrates are included to complete this easy to use staining kit.

Reliable and reproducible results are obtained on tissue sections where the two target antigens do not over-lap (co-localize) in the same structure of the same cell, but rather are expressed in different cell compartments or different cell types.

Also included, BLOXALL Blocking Solution abolishes all endogenous peroxidase and alkaline phosphatase activity in a single 10 minute incubation. In addition, 2.5% Normal Serum blocks all non-specific binding of detection reagents to ensure crisp, specific staining.

Save time and streamline your double staining IHC workflow with the ImmPRESS Duet Double Staining Polymer Kit.  These reagents are blended at optimized dilutions to provide a stable pre-diluted, ready-to-use HRP/AP formula that will detect mouse and rabbit primary antibodies, and facilitate a time-saving, simplified, one-step double label detection protocol. For maximum sensitivity and staining contrast between target antigens, ImmPACT DAB EqV (HRP, brown) and ImmPACT Vector® Red (AP, magenta) substrates are included to complete this easy to use staining kit.

Reliable and reproducible results are obtained on tissue sections where the two target antigens do not over-lap (co-localize) in the same structure of the same cell, but rather are expressed in different cell compartments or different cell types.

Also included, BLOXALL  Blocking Solution abolishes all endogenous peroxidase and alkaline phosphatase activity in a single 10 minute incubation. In addition, 2.5% Normal Serum blocks all non-specific binding of detection reagents to ensure crisp, specific staining.

All reagents are supplied in stabilized, ready-to-use (R.T.U.) solutions for maximum convenience, and consistency between assays. Yes, we conducted studies on the compatibility of the ImmPRESS HRP polymer reagents with three commercially available autostainers (Agilent/Dako Autostainer Plus, Leica Bond Rx, and Ventana Discovery Ultra). We showed that our reagents are suitable for IHC detection on each of these platforms. The ImmPRESS polymer reagents and enzyme substrates generated equivalent IHC staining results compared to reagents from the instrument manufacturer. Some modifications in the protocol were performed to optimize the signal-to-noise ratio, such as a shorter incubation time for the ImmPRESS polymer and increase in the number of buffer washes following polymer incubation. The ImmPRESS-AP Alkaline Phosphatase Polymer Reagent micropolymer technology limits steric interference and provides enhanced accessibility to the target avoiding the disadvantages of other polymer systems that use large dextrans or other macromolecules as backbones.  The result is crisp, strong staining of antibody targets, especially nuclear and membrane antigens, and  greater sensitivity than conventional alkaline phosphatase antibody conjugates and other alkaline phosphatase polymer systems for immunohistochemistry and other applications. 
 

Immunostaining Protocol

The staining procedure is simple as shown in the diagram below. Following a blocking step with the diluted normal horse serum, sections are incubated with primary antibody. After a brief wash, the appropriate ImmPRESS Reagent is added to the sections and incubated for 30 minutes. Sections are again rinsed and the slides are developed with the peroxidase substrate of choice.

Consider Species Cross-Reactivity

When choosing the optimal detection system for your application, it is important to consider not only the species of the primary antibody but also the species of the tissue. If the species of the primary antibody and the species of the tissue are closely related (e.g. rat and mouse), the secondary antibody may bind to endogenous IgG in the tissue section leading to background. The following options minimize background staining in these instances:

  • Use a secondary antibody specifically adsorbed to remove cross-reacting antibodies of closely-related species (e.g. ImmPRESS Anti-Mouse IgG, Rat Adsorbed).
  • Use the M.O.M.® ImmPRESS Kit (Cat. No. MP-2400) for applications of mouse primary antibodies on mouse tissue.

The ImmPRESS Excel Amplified HRP (peroxidase) Polymer Staining Kit is an enzymatic, amplification system that produces crisp, highly sensitive, specific staining with low background. The ImmPRESS Excel Amplified Staining System capitalizes on all the advantages of the ImmPRESS polymerized reporter enzyme staining system technology but offers additional sensitivity and convenience.

This system employs a proprietary ready-to-use (RTU) Amplifier Antibody, followed by a ready-to-use (RTU) ImmPRESS Excel HRP Polymer Reagent. These reagents are affinity purified and extensively cross-adsorbed to ensure high sensitivity and low background. In addition, the ImmPRESS Excel Reagent is conjugated to horseradish peroxidase micropolymers, a novel approach that avoids the use of large dextrans or other macromolecules as a backbone. These unique micropolymers allow a higher density of enzymes per antibody to bind the target with minimal interference. The result is an increase in target accessibility, signal intensity, and significantly less background staining.

The included ImmPACT DAB EqV Substrate produces a crisp, dark brown reaction product with the excellent sensitivity and low background characteristic of the ImmPRESS / ImmPACT combination.
