<p>TMB (3,3′, 5,5′ tetramethylbenzidine dihydrochloride) is a sensitive chromogen substrate for peroxidase and a non-carcinogenic substitute for benzidine. TMB can be used for blotting, immunohistochemical, and <em>in situ</em> hybridization applications. Slides stained with TMB can be dehydrated and permanently mounted and the soluble blue product, produced in the absence of the stabilizing solution, is useful for ELISA assays.</p>
<p>The TMB HRP Substrate Kit (SK-4400) contains the reagents needed to produce either a precipitating blue reaction product or a soluble blue reaction product. This kit also contains a stabilizing solution which, when added to the working solution, allows the development of a blue precipitate. Kit materials are pre-packaged in convenient dropper bottles.</p>

This reagent contains no animal-derived proteins and can be used as an alternative to sera, BSA, casein or non-fat dry milk in situations when animal protein-based blocking solutions cause increased background or interfere with blot development. Primary and secondary antibodies can be diluted in this solution and stored at 4 ºC for up to a week.</td>

		<p>Both products are derived from the same plant protein and in some applications can be used interchangeably. At a high level, one is presented as a ready-to-use (RTU) format and the other a more economical concentrate. The key difference is that the RTU format has been specifically optimized for use in IHC and IF applications. The RTU format has a neutral pH suitable for tissue and cell-based assays and has been more highly refined to avoid the presence of flocculent material that may cause interference with microscopy work. The 5x concentrated format is best suited for membrane blotting applications.</p>

				        <h3>Technical Information</h3><p>This plant protein derived product is a universal antibody diluent and blocking reagent intended for cell and tissue based immunohistochemistry (IHC) and immunofluorescent (IF) applications. This ready-to-use solution can be used as an alternative to normal sera, BSA, casein and non-fat dry milk in methods where these or other blocking agents have been found to be ineffective.</p><p>It is supplied without any sodium azide, and as such, can be used with both peroxidase and alkaline phosphatase antibody conjugates and all secondary detection reagents including polymer systems and avidin/biotin reagents that incorporate these enzymes. </p><p>Importantly, this is a different formulation than our concentrated (5x) animal free blocker, due to the optimized conditions and neutral pH of the solution for IHC and IF methods. </p>                    </div>

		<p>We offer a choice of either peroxidase or alkaline phosphatase based VECTASTAIN ABC kit detection systems. Selection of which enzyme system to use would be the first step. Most IHC applications involve detecting an unconjugated primary antibody. Consider the species in which the primary antibody is raised (e.g. rabbit), and then match to to corresponding species-specific VECTASTAIN ABC kit (e.g. VECTASTAIN ABC Rabbit IgG kit). If you already have a biotinylated target (i.e. biotinylated secondary antibody or primary antibody), then a standard VECTASTAIN ABC kit containing just the avidin/biotinylated enzyme reagents would be required. Note that only the VECTASTAIN Universal Elite PLUS ABC kit (PK-8200) contains a substrate.</p>

For example, to detect a primary antibody made in rabbit, the appropriate choice is a VECTASTAIN ABC Kit designated “Rabbit IgG”.</p><p>For additional versatility and convenience, Vector Laboratories offers secondary antibody products that recognize more than one species of primary antibody. Our Universal Biotinylated Anti-Mouse/Rabbit IgG secondary antibody (Cat. No. BA-1400) is a cocktail of biotinylated anti-mouse IgG and anti-rabbit IgG, designed for use with both rabbit and mouse primary antibodies. This antibody should not be used to stain tissues from rodents or rabbits due to its reactivity with IgG present in the tissues of these species. The Universal Pan-Specific Anti- Mouse/Rabbit/Goat secondary antibody (Cat. 				        <h3>Technical Information</h3><p>The peroxidase micropolymers of the ImmPRESS HRP polymer reagent limit steric interference and provide enhanced accessibility to the target, avoiding the disadvantages of other polymer systems that use large dextrans or other macromolecules as backbones. The result is crisp, strong staining of antibody targets, especially nuclear and membrane antigens (such as Ki67, estrogen receptor, bcl-2, CD3, CD8 and CD10) and greater sensitivity than other polymer systems.</p><h3>Staining Procedure</h3><p>The staining procedure is simple as shown in the diagram below. Following a blocking step with the diluted normal horse serum, sections are incubated with primary antibody. After a brief wash, the appropriate ImmPRESS Reagent is added to the sections and incubated for 30 minutes. 				        <h3>Technical Information</h3><p>The peroxidase micropolymers of the ImmPRESS HRP polymer reagent limit steric interference and provide enhanced accessibility to the target, avoiding the disadvantages of other polymer systems that use large dextrans or other macromolecules as backbones. The result is crisp, strong staining of antibody targets, especially nuclear and membrane antigens (such as Ki67, estrogen receptor, bcl-2, CD3, CD8 and CD10) and greater sensitivity than other polymer systems.</p><h3>Staining Procedure</h3><p>The staining procedure is simple as shown in the diagram below. Following a blocking step with the diluted normal horse serum, sections are incubated with primary antibody. After a brief wash, the appropriate ImmPRESS Reagent is added to the sections and incubated for 30 minutes. If the species of the primary antibody and the species of the tissue are closely related (e.g. rat and mouse), the secondary antibody may bind to endogenous IgG in the tissue section leading to background. The following options minimize background staining in these instances:</p><ul><li>Use a secondary antibody specifically adsorbed to remove cross-reacting antibodies of closely-related species (e.g. ImmPRESS Anti-Mouse IgG, Rat Adsorbed).</li><li>Use the M.O.M.<sup>®</sup> ImmPRESS Kit (Cat. 				        <h3>Technical Information</h3><p>The peroxidase micropolymers of the ImmPRESS HRP polymer reagent limit steric interference and provide enhanced accessibility to the target, avoiding the disadvantages of other polymer systems that use large dextrans or other macromolecules as backbones. The result is crisp, strong staining of antibody targets, especially nuclear and membrane antigens (such as Ki67, estrogen receptor, bcl-2, CD3, CD8 and CD10) and greater sensitivity than other polymer systems.</p><h3>Staining Procedure</h3><p>The staining procedure is simple as shown in the diagram below. Following a blocking step with the diluted normal horse serum, sections are incubated with primary antibody. After a brief wash, the appropriate ImmPRESS Reagent is added to the sections and incubated for 30 minutes. If the species of the primary antibody and the species of the tissue are closely related (e.g. rat and mouse), the secondary antibody may bind to endogenous IgG in the tissue section leading to background. These reagents are blended at optimized dilutions to provide a stable pre-diluted, ready-to-use HRP/AP formula that will detect mouse and rabbit primary antibodies, and facilitate a time-saving, simplified, one-step double label detection protocol. For maximum sensitivity and staining contrast between target antigens, ImmPACT DAB EqV (HRP, brown) and ImmPACT Vector Red (AP, magenta) substrates are included to complete this easy to use staining kit.</p><p>Reliable and reproducible results are obtained on tissue sections where the two target antigens do not over-lap (co-localize) in the same structure of the same cell, but rather are expressed in different cell compartments or different cell types.</p><p>Also included, BLOXALL Blocking Solution abolishes all endogenous peroxidase and alkaline phosphatase activity in a single 10 minute incubation. In addition, 2.5% Normal Serum blocks all non-specific binding of detection reagents to ensure crisp, specific staining.</p><p>All reagents are supplied in stabilized, ready-to-use (R.T.U.) solutions for maximum convenience, and consistency between assays. 				        <h3>Technical Information</h3><p>Save time and streamline your double staining IHC workflow with the ImmPRESS Duet Double Staining Polymer Kit.  These reagents are blended at optimized dilutions to provide a stable pre-diluted, ready-to-use HRP/AP formula that will detect mouse and rabbit primary antibodies, and facilitate a time-saving, simplified, one-step double label detection protocol. For maximum sensitivity and staining contrast between target antigens, ImmPACT DAB EqV (HRP, brown) and ImmPACT Vector<sup>®</sup> Red (AP, magenta) substrates are included to complete this easy to use staining kit.</p><p>Reliable and reproducible results are obtained on tissue sections where the two target antigens do not over-lap (co-localize) in the same structure of the same cell, but rather are expressed in different cell compartments or different cell types.</p><p>Also included, BLOXALL  Blocking Solution abolishes all endogenous peroxidase and alkaline phosphatase activity in a single 10 minute incubation. 		<p>The ImmPRESS Excel kits are a two-step peroxidase polymer based detection system that include an unconjugated amplifier antibody. This intermediate amplifier antibody increases sensitivity of the assay at least three- to four-fold over that of a one-step polymer based system, by facilitating the introduction of more peroxidase enzyme at the site of specific antigen localization. This increase in sensitivity would be advantageous in instances of weak antigen expression, and to further dilute out an expensive primary antibody. The ImmPRESS Excel Amplifier kits are presented as a complete kit format that include enzyme quench, blocking serum and ImmPACT<sup>®</sup> DAB EqV peroxidase substrate, in addition to the amplifier antibody and defined ImmPRESS polymer secondary antibody. However, as the two published references below indicate, this format is still modular and allows for the substitution of different detection reagents. 		<p>Yes, we conducted studies on the compatibility of the ImmPRESS HRP polymer reagents with three commercially available autostainers (Agilent/Dako Autostainer Plus, Leica Bond Rx, and Ventana Discovery Ultra). We showed that our reagents are suitable for IHC detection on each of these platforms. The ImmPRESS polymer reagents and enzyme substrates generated equivalent IHC staining results compared to reagents from the instrument manufacturer. 		<p>The ImmPRESS Excel kits are a two-step peroxidase polymer based detection system that include an unconjugated amplifier antibody. This intermediate amplifier antibody increases sensitivity of the assay at least three- to four-fold over that of a one-step polymer based system, by facilitating the introduction of more peroxidase enzyme at the site of specific antigen localization. This increase in sensitivity would be advantageous in instances of weak antigen expression, and to further dilute out an expensive primary antibody. The ImmPRESS Excel Amplifier kits are presented as a complete kit format that include enzyme quench, blocking serum and ImmPACT<sup>®</sup> DAB EqV peroxidase substrate, in addition to the amplifier antibody and defined ImmPRESS polymer secondary antibody. However, as the two published references below indicate, this format is still modular and allows for the substitution of different detection reagents. 		<p>Yes, we conducted studies on the compatibility of the ImmPRESS HRP polymer reagents with three commercially available autostainers (Agilent/Dako Autostainer Plus, Leica Bond Rx, and Ventana Discovery Ultra). We showed that our reagents are suitable for IHC detection on each of these platforms. The ImmPRESS polymer reagents and enzyme substrates generated equivalent IHC staining results compared to reagents from the instrument manufacturer.
