<p>Metabolic labeling of glycans with unnatural sialic acid analogues has emerged as an invaluable tool for studying sialic acid biology. It has been showed that 9-azido-9-deoxy-N-acetylneuraminic acid (9AzNeu5Ac), a Neu5Ac analogue with the substitution of 9-hydroxyl group with an azide, could be metabolized and incorporated into sialoglycans in living cells and mice. The incorporated sialic acid analogue, 9AzNeu5Ac, enable visualization the cell-surface sialoglycans and glycoproteomic profiling by click-labeling with fluorophores and affinity tags, respectively.</p>

<p>Bis-sulfone NHS Ester is a bis-alkylating labeling reagent that is selective for the cysteine sulfur atoms from a native disulfide. These reagents undergo bis-alkylation to conjugate both thiols derived from the two cysteine residues of a reduced native disulfide bond such as the interchain disulfide bonds of an antibody. The reaction results in covalent rebridging of the disulfide bond via a three carbon bridge leaving the protein structurally intact.</p>
<p>The covalent, site-specific conjugation of PEG to a polyhistidine tag (His-tag) on a protein with bis-alkylating reagents was also reported in literature (see Selected References section).</p>

				        <h3>Description</h3><p>Bis-sulfone-Acid is a bis-alkylating labeling reagent that is selective for the cysteine sulfur atoms from a native disulfide. Bis-alkylating reagents undergo bis-alkylation to conjugate both thiols derived from the two cysteine residues of a reduced native disulfide bond such as the interchain disulfide bonds of an antibody. The reaction results in covalent rebridging of the disulfide bond via a three carbon bridge leaving the protein structurally intact.</p><p>The covalent, site-specific conjugation of PEG to a polyhistidine tag (His-tag) on a protein with bis-alkylating reagents was also reported in literature (see Selected References section).</p>                    </div>

<p>L-Homopropargylglycine (HPG) is an amino acid analog of methionine that contains a very small modification, specifically an alkyne moiety that can be fed to cultured cells and incorporated into proteins during active protein synthesis. Detection utilizes the chemoselective ligation or “click ” reaction between an azide and an alkyne or cyclooctyne. The alkyne-modified protein is detected with either fluorescent azide or biotin azide. Detection sensitivity with these reagents in 1-D gels and Western blots is in the low femtomole range and compatible with downstream LC-MS⁄MS and MALDI MS analysis.</p>
