<p>TCO-NHS Ester is an amine-reactive building block used for modification of amine-containing molecule in organic media. TCO-NHS ester is supplied as a single axial (minor, <em>rel</em>-(1S-2S-5E-pS), ref 1 and 4) diastereomer. Specially formulated crystalline provides easy handling and extended shelf life.</p>
<p>Axial TCO derivatives demonstrate much higher reactivity in the inverse electron demand cycloaddition reaction compared to equatorial TCO derivatives. Up to 10-fold increase in reactivity was reported (ref 4) compared to equatorial TCO derivatives. At the same time, the axial TCO derivatives undergo <em>cis-trans</em> deactivation at faster rate compared to equatorial TCO derivatives (ref 4).</p>

TCO-NHS Ester is not recommended for labeling of proteins in aqueous buffers due to poor level of activation of labeled proteins (<a href="https://pubs.acs.org/doi/10.1021/bc500605g">Rahim, M.K., <em>et al</em>., Bioconjugate Chem. 2015, 26, 352-360</a>). TCO-PEG4-NHS Ester is well suited for protein and antibody labeling in aqueous buffers.</p>

<p>TCO-PEG4-TFP Ester is an amine-reactive, labeling reagent used to modify proteins, antibodies, and other amine-containing biopolymers. The hydrophilic polyethylene glycol (PEG) spacer arm imparts water solubility and provides a long and flexible connection that minimizes steric hindrance involved with ligation to complementary tetrazine-containing molecules.</p>
<p>The N-hydroxysuccinimide (NHS, also known as HOSu) esters of carboxylic acids are the most widely used reactive esters for modifying amine-containing peptides and proteins. NHS esters are notorious for having a short, pH-dependent half-life in aqueous media. A 2,3,5,6-tetrafluorophenol (TFP) is a different type of reactive ester that displays much better stability toward hydrolysis in aqueous media resulting in more efficiency and better reproducible labeling of biopolymers. TFP ester of carboxylic acids react with primary amines at the same rate as NHS ester forming covalent amide bond that is identical to one formed by the reaction between primary amines and NHS esters or sulfo-NHS esters.</p>

<p>TCO-PNB Ester is an amine-reactive building block used for modification of amine-containing molecule in organic media. TCO-PNB Ester is supplied as a single equatorial (major, <em>rel</em>-(1S-2S-5E-pR), ref 1 and 4) diastereomer. Specially formulated crystalline provides easy handling and extended shelf life.</p>
<p>TCO-PNB Ester is not recommended for labeling of proteins in aqueous buffers due to poor level of activation of labeled proteins (Rahim, M.K., et al., Bioconjugate Chem. 2015, 26, 352-360). TCO-PEG4-NHS Ester and TCO-PEG12-NHS Ester are well suited for protein and antibody labeling in aqueous buffers.</p>

<p>0TCO-PEG3-Maleimide enables simple and efficient incorporation of TCO moiety onto antibodies, cysteine-containing peptides and other thiol-containing molecules. The maleimide group specifically and efficiently reacts with reduced thiols (sulfhydryl groups, –SH) at pH 6.5 to 7.5 to form stable thioether bonds. The hydrophilic polyethylene glycol (PEG) spacer arm improves solubility in aqueous buffers.</p>
