The NHS ester (or succinimidyl ester) of this dye is also available for conjugating AZDye™ 350 to the primary amines of proteins or antibodies (Lys), amine-modified oligonucleotides, and other amine-containing molecules.</p>
<p>AZDye™ 350 is a moderately photostable, blue-fluorescent probe optimally excited by the 350 nm laser line routinely used for producing bright, blue fluorescent conjugates for imaging and flow cytometry. The emission of AZDye™ 350 conjugates is slightly shorter than that of AMCA or AMCA conjugates, reducing AZDye™ 350’s spectral overlap with the emission of fluorescein or Alexa Fluor® 488 dye.</p>
<p>AZDye™ 350 dye structurally is identical to Alexa Fluor® Dye 350.</p>

<p>AMCA Acid (7-amino-4-methylcoumarin-3-acetic acid) is one of the most popular blue fluorescent tagging molecules. Often used as contrasting probes for double- and triple-labeling inimmunofluorescence microscopy, arrays and in situ hybridization. The desirable properties of AMCA dyes include a relatively large Stoke’s shift and resistance to photobleaching.</p>
<p>AMCA Acid is a reagent of choice for the preparation of custom activated esters that often are not commercially available. Examples of such activated esters include sulfo-NHS, TFP (2,3,5,6-Tetrafluorophenol), STP (4-Sulfo-2,3,5,6-Tetrafluorophenol, Sodium Salt). Another common application for non-activated carboxylic acid is peptide modification during solid phase synthesis, which usually requires in-situ activation with peptide coupling regents, for example HATU. AMCA Acid is also often used for control experiments, and for calibration.</p>

<p>The carboxylic acid of 5-ROX is used for oligonucleotide labeling and automated DNA sequencing applications. Often a dye of choice for fluorescence resonance energy transfer (FRET) and fluorescence quenching applications. Conjugates of this dye have longer-wavelength spectra than the spectra of Lisaamine™ rhodamine B conjugates, but somewhat shorter-wavelength spectra than those of Texas Red™ conjugates.</p>

				        <h3>Description</h3><p>The carboxylic acid of 5-ROX is used for oligonucleotide labeling and automated DNA sequencing applications. Often a dye of choice for fluorescence resonance energy transfer (FRET) and fluorescence quenching applications. Conjugates of this dye have longer-wavelength spectra than the spectra of Lisaamine™ rhodamine B conjugates, but somewhat shorter-wavelength spectra than those of Texas Red™ conjugates.</p>                    </div>

<p>5(6)-TAMRA Acid (5(6)-Carboxytetramethylrhodamine, TMR, TRITC) is a bright orange-fluorescent dye with excitation ideally suited to the <a href="https://fluoroprobes.com/laser/532-nm/">532 nm</a> or 546 nm laser lines. It has been used widely for preparing peptide, protein, <a href="https://en.wikipedia.org/wiki/Nucleotide" rel="nofollow">nucleotide</a> and nucleic acid conjugates, especially fluorescent antibodies and avidin derivatives used in immunochemistry. The absorbance and emission maxima of TAMRA conjugates are 553 nm and 575 nm respectively.</p>
<p>TAMRA Acid is a reagent of choice for the preparation of custom activated esters that often are not commercially available. Examples of such activated esters include sulfo-NHS, TFP (2,3,5,6-Tetrafluorophenol), STP (4-Sulfo-2,3,5,6-Tetrafluorophenol, Sodium Salt). Another common application for non-activated carboxylic acid is peptide modification during solid phase synthesis, which usually requires in-situ activation with peptide coupling regents, for example HATU. TAMRA Acid is also often used for control experiments, and for calibration.</p>

<p>6-TAMRA Acid (6-Carboxytetramethylrhodamine) is a single, pure isomer of carboxytetramethylrhodamine (TMR) free acid. 6-TAMRA conjugates emit bright orange-fluorescence and its excitation band ideally suited to the 532 nm or 546 nm laser lines. It has been used widely for preparing peptide, protein, nucleotide and nucleic acid conjugates, especially fluorescent antibodies and avidin derivatives used in <a href="https://en.wikipedia.org/wiki/Immunochemistry" rel="nofollow">immunochemistry</a>.</p>
<p>Although the mixed isomers of 5(6)-TAMRA Acid is a preferred, routinely used red fluorescent dye for staining proteins, labeling peptides and nucleotides, the purification of 5(6)-TAMRA labeled peptide and nucleotides might be troublesome due to significant signal broadening in HPLC purification. Peptides and nucleotides labeled with a single isomer TAMRA usually give better resolution in HPLC purification that is often required in the conjugation processes. 5-TAMRA is more often used for labeling peptides and proteins. 6-TAMRA is predominately used for labeling nucleotides and sequencing nucleic acids.</p>

<p>5-TAMRA Acid (5-Carboxytetramethylrhodamine) is a single, pure isomer of carboxytetramethylrhodamine (TMR) free acid. 5-TAMRA conjugates emit bright orange-fluorescence and its excitation band ideally suited to the 532 nm or 546 nm laser lines. It has been used widely for preparing peptide, protein, nucleotide and nucleic acid conjugates, especially fluorescent antibodies and avidin derivatives used in <a href="https://en.wikipedia.org/wiki/Immunochemistry" rel="nofollow">immunochemistry</a>.</p>
<p>Although the mixed isomers of 5(6)-TAMRA Acid is a preferred, routinely used red fluorescent dye for staining proteins, labeling peptides and nucleotides, the purification of 5(6)-TAMRA labeled peptide and nucleotides might be troublesome due to significant signal broadening in HPLC purification. Peptides and nucleotides labeled with a single isomer TAMRA usually give better resolution in HPLC purification that is often required in the conjugation processes. 5-TAMRA is more often used for labeling peptides and proteins. 6-TAMRA is predominately used for labeling nucleotides and sequencing nucleic acids.</p>

Examples of such activated esters include sulfo-NHS, TFP (2,3,5,6-Tetrafluorophenol), and STP (4-Sulfo-2,3,5,6-Tetrafluorophenol, Sodium Salt). Another common application for the non-activated carboxylic acid is peptide modification during solid phase synthesis, which usually requires in-situ activation with peptide coupling regents, e.g. HATU.</p>

<p>AZDye™ 405 Acid is a water-soluble, blue-fluorescent dye that is often used in multi-color applications, including flow cytometry and super-resolution microscopy using STORM. Its excitation is ideally suited for the 407 nm spectral line of the krypton laser or the 408 nm violet laser diode. AZDye™ 405 conjugates are pH-insensitive from pH 4 to pH 10.</p>
<p>The carboxylic acid of AZDye™ 405 is a reagent of choice for the preparation of custom activated esters that often are not commercially available. Examples of such activated esters include sulfo-NHS, TFP (2,3,5,6-Tetrafluorophenol), and STP (4-Sulfo-2,3,5,6-Tetrafluorophenol, Sodium Salt). Another common application for the non-activated carboxylic acid is peptide modification during solid phase synthesis, which usually requires in-situ activation with peptide coupling regents, e.g. HATU. AZDye™ 405 Acid is also often used for control experiments, and for calibration.</p>

<p>MB™ 660R is a bright and photostable far-red dye that emits fluorescence at about 685 nm in the borderline spectral region between far-red and near-IR. Although the absorption maximum is at around 665 nm, this dye can be sufficiently excited by the 633 or 635 nm laser. MB™ 660R dye is water soluble and pH-insensitive from pH 4 to pH 10. MB 660R is a rhodamine-based dye, and like rhodamine dyes in general, it is exceptionally photostable (Figure 1, description tab). The superior photostability and excellent brightness of MB 660R make the dye an ideal choice for confocal microscopy and other demanding applications.</p>

Another common application for the non-activated carboxylic acid is peptide modification during solid phase synthesis, which usually requires in-situ activation with peptide coupling regents, e.g. HATU.</p>
