<p>MB™ 594 is a very hydrophilic, water-soluble red fluorescent dye. Structurally related to Alexa Fluor® 594 dye, MB™ 594 dye contains an additional, negatively charged sulfo group (SO3H) to improve water solubility and minimize self-quenching. Characteristic features of MB™ 594 are strong absorption, high fluorescence quantum yield, high photostability, and pH insensitivity from pH 3 to pH 10. MB™ 594 is a highly suitable dye for single-molecule detection applications and high-resolution microscopy such as PALM, dSTORM, STED etc.</p>
<p><a href="https://en.wikipedia.org/wiki/Maleimide" rel="nofollow">Maleimide</a> is the most popular sulfhydryl-reactive reactive group for conjugating the dye to a thiol group on a protein, oligonucleotide thiophosphate, or low molecular weight ligand. The maleimide group specifically and efficiently reacts with reduced thiols (sulfhydryl groups, –SH) at pH 6.5 to 7.5 to form stable thioether bond. The resulting conjugates exhibit brighter fluorescence and greater photostability than the conjugates of many other spectrally similar fluorophores.</p>

<p>MB™ 488 is a very hydrophilic, water-soluble green emitting dye. Structurally related to Alexa Fluor® 488 dye, MB™ 488 dye is a next step in improving water solubility, brightness and minimizing self-quenching. Characteristic features of the label are strong absorption, high fluorescence quantum yield, and very high photostability. MB™ 488 is a highly suitable dye for single-molecule detection applications and high-resolution microscopy such as PALM, dSTORM, STED etc. Additionally MB™ 488 is an excellent dye for flow cytometry (FACS), fluorescence in-situ hybridization (FISH) and many other applications.</p>
<p>The NHS Ester reacts specifically and efficiently with a primary amine (e.g., side chain of lysine residues or aminosilane-coated surfaces) at pH 7-9 to form a stable, covalent amide bond. The NHS ester (or succinimidyl ester) is the most popular tool for conjugating dyes to the primary amines of protein or antibody (Lys), amine-modified oligonucleotides, and other amine-containing molecules.</p>

<p>MB 594 NHS Ester is a very hydrophilic, water-soluble red fluorescent dye. Structurally related to Alexa Fluor® 594 dye, MB™ 594 dye contains an additional, negatively charged sulfo group (SO3H) to improve water solubility and minimize self-quenching. Characteristic features of MB™ 594 are strong absorption, high fluorescence quantum yield, high photostability, and pH insensitivity from pH 3 to pH 10. MB™ 594 is a highly suitable dye for single-molecule detection applications and high-resolution microscopy such as PALM, dSTORM, STED etc.</p>
<p>The NHS Ester reacts specifically and efficiently with a <a href="http://www.ochempal.org/index.php/alphabetical/o-p/primary-amine/" rel="nofollow">primary amine</a> (e.g., side chain of lysine residues or aminosilane-coated surfaces) at pH 7-9 to form a stable, covalent amide bond. The NHS ester (or succinimidyl ester) is the most popular tool for conjugating dyes to the primary amines of protein or antibody (Lys), amine-modified oligonucleotides, and other amine-containing molecules.</p>

<p><img class="size-full wp-image-5177 alignleft" src="https://staging.vectorlabs.com/wp-content/uploads/2023/09/IR-800-laser.webp" alt="laser line" width="197" height="208" />MB 800Z (also known as s775z) is a conceptually new class of sterically shielded NIR cyanine heptamethine dye that was developed at professor Bradley D. Smith laboratory. This dye contains two shielding PEG arms directly over both faces of the heptamethine fluorochrome blocking any undesired bimolecular association processes and thus dramatically enhances the fluorescence brightness. A recently published antibody labeling study clearly demonstrated 1-2 order of magnitude increase in brightness compared to commercially available NIR dyes as a result of almost complete prevention of stacking of multiple fluorophores appended to the antibody surface (see description section).</p>
<p>MB 800Z is a zwiteroinic, charge-balanced dye with an equal number of anionic sulfonate and cationic ammonium residues, a structural feature that is known to reduce interactions with off -target biological surfaces. MB 800Z will enable researchers to greatly improve various types of indirect NIR immunofluorescence imaging and diagnostics applications that require high sensitivity and also develop new photonintense techniques that require high photostability.</p>
<p>Cyanine heptamethine dyes are well-known NIR fluorophores, with emission wavelengths &#62;740 nm, and often are reagents of choice for labeling antibodies. Two of the most popular commercial NIR cyanine heptamethine dyes for antibody conjugation are <a href="https://www.licor.com/bio/reagents/irdye-800cw-infrared-dyes">IRDye 800CW</a> and <a href="https://www.thermofisher.com/order/catalog/product/46421?us&#38;en#/46421?us&#38;en">DyLight 800</a>. While these NIR dyes are undoubtedly useful for many types of immunofluorescence technologies, the resulting NIR dye-labeled antibodies sometimes exhibit performance limitations due to three inherent fluorophore concerns. (1) A meso-OAryl group connected directly to the heptamethine fluorochrome group is susceptible to nucleophilic displacement by biological amines and thiols<sup>1, 2</sup> resulting in a diminished chemical stability of the dye-antibody conjugates during synthesis, storage, or the time-course of an imaging experiment. In addition, the electron-donating meso-OAryl group promotes high fluorochrome reactivity with electrophilic singlet oxygen, resulting in relatively poor dye photostability.<sup>3,4</sup> (2) Both dyes are flat molecules with a hydrophobic core and a polyanionic charge periphery. Thus, the chemical conversion of a small polar, cationic lysine residue on the antibody surface to a large hydrophobic, polyanionic dye derivative has the potential to produce substantial changes in antibody folding and physiochemical properties, leading to lower antibody stability and decreased target specificity.<sup>4-8</sup> (3) When activated versions of these hydrophobic dyes are conjugated to an antibody surface, they tend to attach at the proximal lysine sites as stacked face-to-face dimers, which produces a diagnostic H-dimer peak in the absorbance spectra that is nonfluorescent.<sup>8,9</sup> Moreover, the close stacking of proximal conjugated IRDye 800CW or DyLight800 on an antibody surface amplifies the potential for a deleterious effect on antibody targeting because of a localized patch of polyanionic charge and hydrophobicity.</p>
<p>In order to address these drawbacks of commercially NIR cyanine a conceptually new class of sterically shielded NIR dyes was developed in Dr. Bradley D. 				        <h3>Description</h3><p><img loading="lazy" decoding="async" class="size-full wp-image-5177 alignleft" src="https://staging.vectorlabs.com/wp-content/uploads/2023/09/IR-800-laser.webp" alt="laser line" width="197" height="208" title="MB 800Z NHS ester 9">MB 800Z (also known as s775z) is a conceptually new class of sterically shielded NIR cyanine heptamethine dye that was developed at professor Bradley D. Smith laboratory. This dye contains two shielding PEG arms directly over both faces of the heptamethine fluorochrome blocking any undesired bimolecular association processes and thus dramatically enhances the fluorescence brightness. A recently published antibody labeling study clearly demonstrated 1-2 order of magnitude increase in brightness compared to commercially available NIR dyes as a result of almost complete prevention of stacking of multiple fluorophores appended to the antibody surface (see description section).</p><p>MB 800Z is a zwiteroinic, charge-balanced dye with an equal number of anionic sulfonate and cationic ammonium residues, a structural feature that is known to reduce interactions with off -target biological surfaces. MB 800Z will enable researchers to greatly improve various types of indirect NIR immunofluorescence imaging and diagnostics applications that require high sensitivity and also develop new photonintense techniques that require high photostability.</p><p>Cyanine heptamethine dyes are well-known NIR fluorophores, with emission wavelengths &gt;740 nm, and often are reagents of choice for labeling antibodies. Two of the most popular commercial NIR cyanine heptamethine dyes for antibody conjugation are <a href="https://www.licor.com/bio/reagents/irdye-800cw-infrared-dyes" target="_blank" rel="noopener">IRDye 800CW</a> and <a href="https://www.thermofisher.com/order/catalog/product/46421?us&amp;en#/46421?us&amp;en" target="_blank" rel="noopener">DyLight 800</a>. While these NIR dyes are undoubtedly useful for many types of immunofluorescence technologies, the resulting NIR dye-labeled antibodies sometimes exhibit performance limitations due to three inherent fluorophore concerns. (1) A meso-OAryl group connected directly to the heptamethine fluorochrome group is susceptible to nucleophilic displacement by biological amines and thiols<sup>1, 2</sup> resulting in a diminished chemical stability of the dye-antibody conjugates during synthesis, storage, or the time-course of an imaging experiment. In addition, the electron-donating meso-OAryl group promotes high fluorochrome reactivity with electrophilic singlet oxygen, resulting in relatively poor dye photostability.<sup>3,4</sup> (2) Both dyes are flat molecules with a hydrophobic core and a polyanionic charge periphery. Thus, the chemical conversion of a small polar, cationic lysine residue on the antibody surface to a large hydrophobic, polyanionic dye derivative has the potential to produce substantial changes in antibody folding and physiochemical properties, leading to lower antibody stability and decreased target specificity.<sup>4-8</sup> (3) When activated versions of these hydrophobic dyes are conjugated to an antibody surface, they tend to attach at the proximal lysine sites as stacked face-to-face dimers, which produces a diagnostic H-dimer peak in the absorbance spectra that is nonfluorescent.<sup>8,9</sup> Moreover, the close stacking of proximal conjugated IRDye 800CW or DyLight800 on an antibody surface amplifies the potential for a deleterious effect on antibody targeting because of a localized patch of polyanionic charge and hydrophobicity.</p><p>In order to address these drawbacks of commercially NIR cyanine a conceptually new class of sterically shielded NIR dyes was developed in Dr. Bradley D. <p>For many years, the cyanine-5 based dye (Cy™5 and more recently Alexa Fluor® 647) has been the dye of choice in far-red spectral window due to minimal autofluorescence in most biological samples in this spectral region allowing for extremely sensitive detection. In spite of recent improvement in cyanine-5 based dyes most dye are still lacks sufficient photostability required for many demanding applications.</p>

Although the absorption maximum is at around 665 nm, this dye can be sufficiently excited by the 633 or 635 nm laser. MB™ 660R dye is water soluble and pH-insensitive from pH 4 to pH 10. MB 660R is a rhodamine-based dye, and like rhodamine dyes in general, it is exceptionally photostable (Figure 1, description tab). The superior photostability and excellent brightness of MB 660R make the dye an ideal choice for confocal microscopy and other demanding applications.</p>
<p>TFP (tetrafluorophenyl) ester is an amine-reactive activated ester that reacts with primary amines of biomolecules in the same way as the succinimidyl ester (SE or NHS-ester) to form a stable amide bond. The major advantage of TFP esters over the succinimidyl ester is better resistance to spontaneous hydrolysis during conjugation reactions, which results in more efficient and reproducible labeling of biopolymers. TFP esters are stable for several hours at the basic pH typically used for reactions–far outlasting succinimidyl esters.</p>

TFP esters are stable for several hours at the basic pH typically used for reactions–far outlasting succinimidyl esters.</p><p>TFP (tetrafluorophenyl) ester is an amine-reactive activated ester that reacts with primary amines of biomolecules in the same way as the succinimidyl ester (SE or NHS-ester) to form a stable amide bond. The major advantage of TFP esters over the succinimidyl ester is better resistance to spontaneous hydrolysis during conjugation reactions, which results in more efficient and reproducible labeling of biopolymers. Although the absorption maximum is at around 680 nm, this dye can be sufficiently excited by the 633 or 635 nm laser. MB™ 680R dye is water soluble and pH-insensitive from pH 4 to pH 10. MB 680R is a rhodamine-based dye, and like rhodamine dyes in general, it is exceptionally photostable. The superior photostability and excellent brightness of MB 680R make the dye an ideal choice for confocal microscopy and other demanding applications.</p><p>The NHS Ester reacts specifically and efficiently with a primary amine (e.g., side chain of lysine residues or aminosilane-coated surfaces) at pH 7-9 to form a stable, covalent amide bond. The NHS ester (or succinimidyl ester) is the most popular tool for conjugating dyes to the primary amines of protein or antibody (Lys), amine-modified oligonucleotides, and other amine-containing molecules.</p></div>                    </div>

<p>TFP (tetrafluorophenyl) ester is an amine-reactive activated ester that reacts with primary amines of biomolecules in the same way as the succinimidyl ester (SE or NHS-ester) to form a stable amide bond. The major advantage of TFP esters over the succinimidyl ester is better resistance to spontaneous hydrolysis during conjugation reactions, which results in more efficient and reproducible labeling of biopolymers. TFP esters are stable for several hours at the basic pH typically used for reactions–far outlasting succinimidyl esters.</p>
