Both products are derived from the same plant protein and in some applications can be used interchangeably. At a high level, one is presented as a ready-to-use (RTU) format and the other a more economical concentrate. The key difference is that the RTU format has been specifically optimized for use in IHC and IF applications. The RTU format has a neutral pH suitable for tissue and cell-based assays and has been more highly refined to avoid the presence of flocculent material that may cause interference with microscopy work. The 5x concentrated format is best suited for membrane blotting applications.

Technical Information

This plant protein derived product is a universal antibody diluent and blocking reagent intended for cell and tissue based immunohistochemistry (IHC) and immunofluorescent (IF) applications. This ready-to-use solution can be used as an alternative to normal sera, BSA, casein and non-fat dry milk in methods where these or other blocking agents have been found to be ineffective.

It is supplied without any sodium azide, and as such, can be used with both peroxidase and alkaline phosphatase antibody conjugates and all secondary detection reagents including polymer systems and avidin/biotin reagents that incorporate these enzymes. 

Importantly, this is a different formulation than our concentrated (5x) animal free blocker, due to the optimized conditions and neutral pH of the solution for IHC and IF methods.

Technical Information

To maximize the signal to noise ratio, the correct dilutions of primary antibody and secondary antibody conjugate need to be empirically determined and depend on several factors including the membrane type, the substrate to be used, the blocking/diluent solution and the amount of target. WestVision™ Block and Diluent is intended to be used as supplied without further dilution.

Due to the high sensitivity provided by the WestVision™ Block and Diluent, lower concentration of primary antibody and secondary antibody conjugates may be required to achieve optimal results.

As a starting point for chemiluminescent detection, dilute the primary antibody to 0.1 to 1.0 ug/ml and dilute the secondary antibody conjugate to 0.02 – 0.2 ug/ml, in WestVision™ Block and Diluent. 

As a starting point for chromogenic detection, dilute the primary antibody to 0.25 to 1.0 ug/ml and dilute the secondary antibody conjugate to 0.2 – 1.0 ug/ml in WestVision™ Block and Diluent.

IMPORTANT: Not all blocking solutions are appropriate for all Western blot assays. Blocking solutions should be tested for compatibility in a given assay. 

PROCEDURE

  1. Remove the membrane from the transfer device.

  2. Rinse briefly with deionized (DI) H2O.

  3. Add enough WestVision™ Block and Diluent to completely cover the membrane and incubate with agitation for 30 – 60 minutes.

  4. Continue with your detection protocol using WestVision™ Block and Diluent to dilute primary antibody and detection reagents.

These kits were developed to prevent non-specific binding of avidin or streptavidin based detection reagents primarily to endogenous biotin in cell and tissue IHC and IF applications. It is generally recommended to match the blocking and detection reagent such as using an avidin/biotin blocking kit with an avidin based detection system. In most cases, however, these kits can be used interchangeably, except in circumstances where avidin or streptavidin may bind to inherent non-biotin associated structures (see: Alon, R., et al {1992}, Eur. J. Cell Biol. 58:271-279).

If the primary antibody is unconjugated, then the avidin/biotin blocking kit reagents can be applied at any time prior to the application of the biotinylated secondary antibody. If the primary antibody is biotinylated, then we would recommend applying the avidin/biotin blocking kit reagents before primary antibody incubation.
