Description

Alkyne Sphinganine a metabolic labeling reagent that contains a terminal alkyne moiety that allows for the subsequent tagging of protein−lipid complexes by click chemistry. The terminal alkyne group can be used in a highly specific linking reaction with azide-containing reagents, known as ‘click chemistry’, in the presence of a copper (Cu)-containing catalyst allowing for the visualization and proteomic profiling of protein−sphingolipid complexes in living cells.

Description

This reagent is a modified lipid containing an omega-terminal alkyne. The terminal alkyne group can be used in a highly specific linking reaction with azide-containing reagents, known as ‘click chemistry’, in the presence of a copper (Cu)-containing catalyst.

E-Cholesterol Alkyne (19-ethynylcholesterol) represents a versatile, sensitive, and easy-to-use tool for tracking cellular cholesterol metabolism and localization as it allows for manifold detection methods including mass spectrometry, and fluorescence microscopy.

E-cholesterol Alkyne (19-ethynylcholesterol) is accepted by cellular enzymes from different biological species (Brevibacterium, yeast, rat, human) and these enzymes include cholesterol oxidases, hydroxylases, and acyl transferases that generate the expected metabolites in in vitro and in vivo assays. Using fluorescence microscopy, researchers can study the distribution of cholesterol at subcellular resolution, detecting the lipid in the Golgi and at the plasma membrane, but also in the endoplasmic reticulum and mitochondria.

In summary, alkyne cholesterol represents a versatile, sensitive, and easy-to-use tool for tracking cellular
cholesterol metabolism and localization as it allows for manifold detection methods including mass spectrometry, and fluorescence microscopy.

Description

This reagent is a modified lipid containing an omega-terminal alkyne. The terminal alkyne group can be used in a highly specific linking reaction with azide-containing reagents, known as ‘click chemistry’, in the presence of a copper-containing catalyst.

Alkyne cholesterol represents a versatile, sensitive, and easy-to-use tool for tracking cellular cholesterol metabolism and localization as it allows for manifold detection methods including mass spectrometry, and fluorescence microscopy.

27-Alkyne cholesterol is accepted by cellular enzymes from different biological species (Brevibacterium, yeast, rat, human) and these enzymes include cholesterol oxidases, hydroxylases, and acyl transferases that generate the expected metabolites in in vitro and in vivo assays.

Description

Alkynyl stearic acid is can be used to identify and characterize the post-translational acylation of proteins using the powerful click chemistry, a simple and robust two-step labeling and detection technique. Due to the very small size of the alkyne group and its complete absence in any natural molecules the presence of alkynyl group does not affect the biodistribution of alkynyl palmitic acid in comparison to the natural palmitic acid making it a suitable substrate for enzymes.
