					<description><![CDATA[<p>Biotin-dPEG®12-DBCO, product number QBD-11811, is a click chemistry biotinylation reagent built on a medium-length (40 atoms, 46.3 Å) single molecular weight PEG linker with a discrete chain length (dPEG®) and functionalized with dibenzylcyclooctyne (DBCO) for strain-promoted azide-alkyne cycloaddition (SPAAC), also known as "copper-free click chemistry." The DBCO functional group reacts with any available azide moiety on a target molecule via SPAAC. <p>Biotinylation is the process of covalently attaching biotin to a molecule such as a peptide, protein, or nucleic acid, or surfaces such as glass or gold. Bioconjugation research and product development and numerous clinical assays use biotinylated molecules. The amphiphilic dPEG® linker of Biotin-dPEG®11-oxyamine·HCl imparts excellent water solubility to biotin, which is usually poorly soluble in water and causes non-specific hydrophobic interactions when conjugated to biomolecules.</p>
<p>Some commercially available biotin-aminooxy products use either no spacer or a hydrophobic alkyl chain as a spacer between biotin and the aminooxy moiety. Such products have poor solubility in water and must be dissolved in a dry organic solvent such as dimethyl sulfoxide (DMSO) immediately before use, which can be problematic under some conditions. If used on proteins, these hydrophobic products may trigger aggregation and precipitation of the biotinylated molecule. Even if aggregation and precipitation do not occur, the hydrophobic spacer can contribute to non-specific binding. Because it is amphiphilic, Biotin-dPEG®11-oxyamine·HCl can be dissolved in water or aqueous buffer and used directly in a conjugation reaction. QBD-11100 will not cause aggregation or precipitation of the biotinylated molecule. Moreover, the hydrophilicity of the dPEG® linker reduces or eliminates non-specific binding.</p>
<p>An aminooxy group (also known as an alkoxyamine) consists of a primary terminal amine adjacent to oxygen (i.e., —ONH2). This group reacts selectively and efficiently with aldehydes to form an aldoxime linkage and with ketones to form a ketoxime linkage. An oxime bond is more stable than a hydrazone bond and does not need to be reduced to a secondary amine to stabilize it. Aldehydes and ketones occur relatively rarely in nature; however, using sodium periodate, aldehydes can be formed in glycoproteins whose carbohydrate coats contain reducing sugars. Moreover, scientific literature examples demonstrate the introduction of ketones into bacterial cell walls through liposomal fusion.</p>

					<description><![CDATA[<p>m-dPEG®25-amido-dPEG®24-DSPE, product number QBD-11095, modifies the lipid 1,2-Distearoyl-sn-glycero-3-phosphoethanolamine (DSPE) with a long (153 atoms, 179.8 Å), methoxy-terminated, single molecular weight, discrete polyethylene glycol (dPEG®) spacer. The spacer length is equivalent to a monodispersed mPEG49. This product is designed to protect liposomes and micelles from opsonization and elimination by the reticuloendothelial system (RES). The m-dPEG® spacer offers protection from opsonization comparable to the traditional polymer mPEG2000, but the monodispersed m-dPEG® product is a single compound, not a non-uniform (dispersed) polymer. The methyl-capped dPEG® spacer has a neutral charge.</p>
<p>Vector Laboratories' dPEG® products are highly pure single molecular weight compounds with a discrete chain length. Traditional PEG compounds used to coat liposomes and micelles consist of an intractable mixture of different chain lengths and molecular weights. The non-uniform polymeric mixture leads to irreproducible purity profiles, unlike dPEG® compounds.</p>

				        <h3>Description</h3><p>Biotin-dPEG®3-oxyamine·HCl, product number QBD-11100, consists of a short (14 atoms, 16.6 Å) single molecular weight, discrete PEG (dPEG®) spacer functionalized at each end with biotin and an oxyamine group (as the HCl salt), respctively. This PEG biotin compound reacts with aldehydes and ketones to forms stable oxime bonds. Using this biotinylation reagent permits biotin labeling of the carbohydrate coats of glycoproteins after the introduction of aldehydes and ketones on the carbohydrate coat using enzymatic (for example, galactose oxidase) or mild chemical reaction conditions such as sodium periodate.</p><p>Biotinylation is the process of covalently attaching biotin to a molecule such as a peptide, protein, or nucleic acid, or surfaces such as glass or gold. Bioconjugation research and product development and numerous clinical assays use biotinylated molecules. The amphiphilic dPEG® linker of Biotin-dPEG®3-oxyamine·HCl imparts excellent water solubility to biotin, which is usually poorly soluble in water and causes non-specific hydrophobic interactions when conjugated to biomolecules.</p><p>Some commercially available biotin-aminooxy products use either no spacer or a hydrophobic alkyl chain as a spacer between biotin and the aminooxy moiety. Such products have poor solubility in water and must be dissolved in a dry organic solvent such as dimethyl sulfoxide (DMSO) immediately before use, which can be problematic under some conditions. If used on proteins, these hydrophobic products may trigger aggregation and precipitation of the biotinylated molecule. Even if aggregation and precipitation do not occur, the hydrophobic spacer can contribute to non-specific binding. Because it is amphiphilic, Biotin-dPEG®3-oxyamine·HCl can be dissolved in water or aqueous buffer and used directly in a conjugation reaction. QBD-11100 will not cause aggregation or precipitation of the biotinylated molecule. Moreover, the hydrophilicity of the dPEG® linker reduces or eliminates non-specific binding.</p><p>An aminooxy group (also known as an alkoxyamine) consists of a primary terminal amine adjacent to oxygen (i.e., —ONH2). This group reacts selectively and efficiently with aldehydes to form an aldoxime linkage and with ketones to form a ketoxime linkage. An oxime bond is more stable than a hydrazone bond and does not need to be reduced to a secondary amine to stabilize it. Aldehydes and ketones occur relatively rarely in nature; however, using sodium periodate, aldehydes can be formed in glycoproteins whose carbohydrate coats contain reducing sugars. Moreover, scientific literature examples demonstrate the introduction of ketones into bacterial cell walls through liposomal fusion.</p>                    </div>

					<description><![CDATA[<p>Biotin-dPEG®7-NH2 (Biotin-dPEG®7-amine), product number QBD-10826, is a short (25 atoms, 29.8 Å), hydrophilic, water-soluble, discrete PEG biotinylation product that reacts with carboxylic acids and their activated esters [N-hydroxysuccinimidyl (NHS), 2,3,5,6-tetrafluorophenyl (TFP)] to form stable amide bonds. Additionally, Biotin-dPEG®7-NH2 forms amide bonds directly with carboxylic acids using 1-Ethyl-3-(3-dimethylaminopropyl)carbodiimide (EDC) chemistry. Furthermore, the product can react with carbonyl groups (aldehydes or ketones) to form a labile Schiff base, which can be reduced to a secondary amine using sodium cyanoborohydride for additional conjugate stability.</p>
<p>Amphiphilic biotin-dPEG®7-NH2 dissolves equally well in both aqueous buffer and organic solvent. It is not necessary to dissolve this product in an organic solvent before using it in aqueous reaction media. Furthermore, biotinylation of biomacromolecules with QBD-10826 will not cause aggregation and precipitation. Eliminating aggregation should consequently improve the signal-to-noise ratio of applications developed with this product. Aldehydes are uncommon in biomacromolecules. However, the oxidation of reducing sugars in glycosylated proteins (for example, using sodium periodate) forms aldehydes on the carbohydrate coats of glycoproteins. Reacting Biotin-dPEG®7-amine with aldehydes will produce labile imines or Schiff bases that can be reduced under mild conditions with sodium cyanoborohydride to form more stable secondary amines.</p>
<p>Any application that can take advantage of the strong biotin-avidin/ streptavidin affinity can use Biotin-dPEG®7-NH2. The terminal amine of QBD-10826 offers the opportunity to biotinylate (1) the carbohydrate coat of glycoproteins and (2) surfaces consisting of carboxylic acids or their active esters. Thus, Biotin-dPEG®7-NH2 expands the range of molecules and nanoparticles for which dPEG®-biotin is useful.</p>

Instead, the well-defined dPEG® product will yield predictable, identifiable conjugates. When using Biotin-dPEG®7-azide with proteins or peptides engineered to contain alkyne side chains at specific locations, both the number and location of conjugation sites are predictable.</p>
<p>As a biotinylation reagent, Biotin-dPEG®7-azide works in various applications. Pull-down assays, affinity purification, plate-type assays such as ELISA, and supramolecular construction can all incorporate Biotin-dPEG®7-azide.</p>

					<description><![CDATA[<p>Biotin-dPEG®23-azide, product number QBD-10787, is a water-soluble biotinylation product functionalized with azide for click chemistry use, on a long (73 atoms) single molecular weight, discrete PEG (dPEG®) chain. This product enables biotinylation of molecules using either metal-catalyzed (CuAAC, RuAAC) or strain-promoted azide-alkyne cycloaddition (SPAAC) chemistry. Consequently, these products allow new types of biotinylated constructs.</p>
<p>Because Biotin-dPEG®23-azide is a single molecular weight dPEG® product, analysis of the resulting conjugates is simplified. The researcher using this product does not have to analyze a variety of PEG chain lengths and molecular weights. Instead, the well-defined dPEG® product will yield predictable, identifiable conjugates. When using Biotin-dPEG®23-azide with proteins or peptides engineered to contain alkyne side chains at specific locations, both the number and location of conjugation sites are predictable.</p>
<p>Biotin-dPEG®23-azide has numerous uses. Through protein engineering, non-natural amino acids with alkyne side chains can be incorporated into distinct locations on proteins, thereby enabling site-specific, reproducible biotinylation of proteins. Likewise, synthetic peptides can be constructed that allow azide-alkyne click chemistry-driven biotinylation of the peptides. Also, surfaces functionalized with alkyne groups can be biotinylated using click chemistry. Furthermore, this product can be modified with other products from Vector Laboratories to create new compounds tailored to the customer’s specific needs. For example, in a 2018 study published in the journal Langmuir, researchers from The BioDesign Institute at Arizona State University used Biotin-dPEG®23-azide to construct a Y-shaped, bivalent, bis-biotin structure with a rigid core to probe the interaction of biotin and streptavidin using both atomic force microscopy (AFM) and surface plasmon resonance (SPR).</p>
<p>As a biotinylation reagent, Biotin-dPEG®23-azide functions like other biotinylation reagents. For example, pull-down assays, affinity purification, ELISA and other plate-type assays, and supramolecular construction can employ Biotin-dPEG®23-azide successfully. Many other applications using Biotin-dPEG®23-azide are possible.</p>

					<description><![CDATA[<p>Biotin-dPEG®23-NH2 (Biotin-dPEG®23-amine), product number QBD-10786, is a carboxylate-reactive biotinylation product on a long (71 atoms), single molecular weight, discrete PEG (dPEG®) spacer. Biotin-dPEG®23-NH2 reacts with activated esters (N-hydroxysuccinimide, 2,3,5,6-tetrafluorophenyl) of carboxylic acids under slightly basic conditions. Moreover, it can be coupled directly to a carboxylic acid using 1-Ethyl-3-(3-dimethylaminopropyl)carbodiimide (EDC) chemistry. Additionally, the product reacts with aldehydes or ketones to form a labile Schiff base. The Schiff base can be reduced to a secondary amine using sodium cyanoborohydride if additional conjugate stability is necessary.</p>
<p>Amphiphilic Biotin-dPEG®23-NH2 dissolves equally well in both aqueous media and organic solvents. Therefore, it is unnecessary to dissolve this product in an organic solvent before using it in an aqueous reaction. Furthermore, biotinylation of biomacromolecules with Biotin-dPEG®23-NH2 will not cause aggregation and precipitation. Eliminating aggregation and precipitation improves the signal-to-noise ratio of applications developed with this product. Reactions with carboxylic acids or active esters of carboxylic acids yield stable amide bonds. Aldehydes are not commonly found in biomacromolecules but can be formed by oxidizing reducing sugars in glycosylated proteins. Reactions with aldehydes create labile imines (Schiff bases) that can be reduced under mild conditions with sodium cyanoborohydride to form more stable secondary amines.</p>
<p>Biotin-dPEG®23-NH2 works in any application that can take advantage of the exceptionally strong biotin-avidin/streptavidin affinity. The terminal amine of QBD-10786 offers the opportunity (1) to biotinylate the carbohydrate coat of glycoproteins by oxidizing reducing sugars to aldehydes and (2) to biotinylate surfaces consisting of carboxylic acids or their active esters. Thus, this product expands the range of molecules and nanoparticles in which dPEG®-biotin can be employed.</p>

Moreover, it can be coupled directly to a carboxylic acid using 1-Ethyl-3-(3-dimethylaminopropyl)carbodiimide (EDC) chemistry. Additionally, the product reacts with aldehydes or ketones to form a labile Schiff base. The Schiff base can be reduced to a secondary amine using sodium cyanoborohydride if additional conjugate stability is necessary.<br />Amphiphilic Biotin-dPEG®23-NH2 dissolves equally well in both aqueous media and organic solvents. Therefore, it is unnecessary to dissolve this product in an organic solvent before using it in an aqueous reaction. Furthermore, biotinylation of biomacromolecules with Biotin-dPEG®23-NH2 will not cause aggregation and precipitation. Eliminating aggregation and precipitation improves the signal-to-noise ratio of applications developed with this product. Reactions with carboxylic acids or active esters of carboxylic acids yield stable amide bonds. Aldehydes are not commonly found in biomacromolecules but can be formed by oxidizing reducing sugars in glycosylated proteins. Reactions with aldehydes create labile imines (Schiff bases) that can be reduced under mild conditions with sodium cyanoborohydride to form more stable secondary amines.</p><p>Biotin-dPEG®23-NH2 works in any application that can take advantage of the exceptionally strong biotin-avidin/streptavidin affinity. The terminal amine of QBD-10786 offers the opportunity (1) to biotinylate the carbohydrate coat of glycoproteins by oxidizing reducing sugars to aldehydes and (2) to biotinylate surfaces consisting of carboxylic acids or their active esters. Thus, this product expands the range of molecules and nanoparticles in which dPEG®-biotin can be employed.</p>                    </div>
