For QBD-10201, the linker is a dPEG®3 product. The flexible, short-chain, discrete PEG (dPEG®) linker imparts water solubility to conjugates formed with it. The maleimide moiety is a popular reactive group for conjugating to free thiol groups in biomolecules and other sulfhydryl compounds. Maleimide groups can be conjugated to free thiol groups in the pH range 6.5 – 7.5, forming a stable thioether linkage.</p>
<p>This product has proven helpful in various applications that utilize the high streptavidin-biotin binding affinity. Such applications include<br />

<p>To use Biotin-dPEG®3-MAL, dissolve the product in a dry solvent and add it to an aqueous compound containing the free thiol. The water-miscible solvent N,N’-dimethylacetamide (DMAC) dried over 3Å molecular sieves is an excellent solvent for this purpose. For biomolecules (e.g., antibodies), care should be taken to use a minimal amount of organic solvent so as not to denature the biomolecule or cause salt precipitation."</p>

<h3>Description</h3>
<p>Biotin-dPEG®11-MAL, product number QBD-10195, is one of three products containing a biotin on one end of a dPEG® linker and a maleimidopropyl moiety on the other end. The maleimide group is a useful, highly popular reactive group for conjugating to free thiol groups in biomolecules. This product provides a short-chain, single molecular weight, discrete PEG (dPEG®) spacer between the biotin and the maleimide. Maleimide groups can be conjugated to free thiol groups in the pH range from 6.5 – 7.5, forming a stable thioether linkage. The dPEG® spacer in this product increases the hydrophilicity of the conjugate.</p>
<p>This product has proven useful in a variety of applications that take advantage of the high streptavidin-biotin binding affinity. Such applications include magnetic bead-based assays, assays to quantitate reducible thiols in proteins, atomic force microscopy, and nanoparticle applications, among others.</p>
<p>To use Biotin-dPEG®11-MAL, the product should be dissolved in dry solvent and then added to an aqueous solution of the compound containing the free thiol. The water-miscible solvent N,N’-dimethylacetamide (DMAC) dried over 3Å molecular sieves is an excellent solvent to use for this purpose. For biomolecules (e.g., antibodies), care should be taken to keep the amount of organic solvent to a minimum so as not to denature the biomolecule or cause salt precipitation.</p>

ChromaLINK Biotin Maleimide has been engineered to include many novel features. As illustrated in Figure 1, the molecule’s structure contains a bis-aryl hydrazone chromophore (a), linked by a PEG3 linker arm (b), to biotin (c). This reagent permits direct spectroscopic quantification of incorporated biotin. Instead, biotin incorporation is quantified by means of a simple spectrophotometric measurement at two wavelengths (A280 / A354). Typical labeling results are illustrated in Figure 2 by spectral overlay scans of four samples. As illustrated, bovine Serum Albumin (100 ul @ 1 mg/ml) was labeled at 0, 5, 10, and 20 mole equivalents using ChromaLINK Biotin Maleimide. <p>Biotin (Long Arm) Maleimide is designed for biotinylating proteins, nucleic acids or other molecules containing one or more thiol groups. The presence of the 6-aminohexanoate spacer arm between the maleimide group and biotin reduces the possibility of steric hinderance. Molecules to be labeled with Biotin (Long Arm) Maleimide require free thiol groups. In the case of proteins, Biotin (Long Arm) Maleimide will react with exposed cysteine residues. Alternatively, free thiols may be generated by reducing disulfide bonds or by modifying other reactive groups such as primary amines with compounds like Traut’s reagent (2-iminothiolane).</p>

				        <h3>Description</h3><p>Biotin (Long Arm) Maleimide is designed for biotinylating proteins, nucleic acids or other molecules containing one or more thiol groups. The presence of the 6-aminohexanoate spacer arm between the maleimide group and biotin reduces the possibility of steric hinderance. Molecules to be labeled with Biotin (Long Arm) Maleimide require free thiol groups. In the case of proteins, Biotin (Long Arm) Maleimide will react with exposed cysteine residues. Alternatively, free thiols may be generated by reducing disulfide bonds or by modifying other reactive groups such as primary amines with compounds like Traut&#8217;s reagent (2-iminothiolane).</p>                    </div>
